摘要:骨质疏松症(osteoporosis)是由于多种原因导致的骨密度下降,以骨量减少、骨微结构破坏为特征的全身性骨病。其发病机制并未完全阐明,目前研究聚焦在微小RNA(microRNA,miRNA)通过调控靶基因表达而影响骨质疏松症的发生和发展,这也成为该领域的研究热点。miRNA作为一类单链非编码小分子,参与细胞的生长、发育及增殖、凋亡等过程。已有研究提示miRNA有可能通过靶基因Runt相关转录因子2(Runt-related transcription factor 2,Runx2)调节成骨细胞的分化成熟,从而影响骨质疏松的发生发展。本文对miRNA介导靶基因Runx2调控成骨细胞分化的分子机制进行综述,以期为骨质疏松症的防治提供新思路。
Osteoporosis is a systemic bone disease characterized by decreased bone mass and destruction of bone microstructure due to a variety of causes.The pathogenesis of osteoporosis has not been fully clarified.Current studies focus on the effect of microRNA(miRNA)on the occurrence and development of osteoporosis by regulating the expression of target genes,which has become a research hotspot in this field.As a class of single-chain non-coding small molecules,miRNA is involved in the growth,development,proliferation,apoptosis,and other processes of cells.Previous studies have suggested that miRNA may regulate the differentiation and maturation of osteoblasts through the target gene Runx2,thus affecting the occurrence and development of osteoporosis.This article summarizes the molecular regulation mechanism of miRNA through mediating target gene Runx2 to regulate osteoblast differentiation,in order to provide a new way of thinking for the prevention and treatment of osteoporosis.
