摘要:目的 基于数据挖掘技术探析中药治疗肝肾亏虚型腰椎间盘突出症(lumbar disc herniation,LDH)的用药规律,运用网络药理学研究核心中药治疗肝肾亏虚型LDH的作用机制。方法 检索中国知网、维普、万方和中国生物医学文献数据库,将纳入的中药进行频数统计、关联规则、复杂网络及聚类分析;运用TCMSP数据库、Uniprot数据库搜索核心中药的成分和靶点,与GeneCards数据库等平台获得的疾病靶点取交集,通过cytoscape3.9.1构建复杂网络、STRING平台完成PPI网络、R语言进行基因本体论(GO)和京都基因与基因组百科全书(KEEG)通路富集分析。结果 检索得到处方53首,中药138味,性味以甘、温为主,归肝、肾二经;其中补虚药占比最高,其次为祛风湿药,功效主要是补益肝肾;二项关联规则和三项关联规则共40条,聚类分析分为5类。核心中药的关键靶点共10个;GO分析获得细胞生物过程(BP)127个、细胞组成(CC)10个、分子功能(MF)20个,KEEG通路127条,选前20条做可视化处理。结论 中药治疗肝肾亏虚型LDH以补益肝肾为核心治法,辅以补气活血、除痹止痛;以补虚药为主,常与祛风湿、通经络药物配合使用。核心中药治疗肝肾亏虚型LDH以多靶点、多通路、多途径为特点。
Objective To explore the medication rules of traditional Chinese medicine in the treatment of lumbar disc herniation(LDH)with liver and kidney deficiency based on data mining technology,and use network pharmacology to study the mechanism of core traditional Chinese medicine in the treatment of liver and kidney deficiency type LDH.Methods CNKI,VIP,WanFang and Chinese Biomedical Literature Database were searched,and frequency statistics,association rules,complex network and cluster analysis were performed on the included TCMs;TCMSP and Uniprot were used to search for the components and targets of core TCMs.The disease targets obtained from platforms such as GeneCards were intersected,and complex networks were constructed by cytoscape3.9.1,PPI network was completed by STRING platform,and GO and KEEG pathway enrichment analysis was performed by R language.Results 53 prescriptions were retrieved,138 traditional Chinese medicines were found.The nature and flavor were mainly sweet and warm,and they mainly belonged to the two channels of liver and kidney.Among them,tonic drugs accounted for the highest proportion,followed by rheumatism drugs,which mainly benefited liver and kidney.There were a total of 40 association rules and three association rules,and the cluster analysis was divided into 5 categories.There were 10 key targets of core traditional Chinese medicines;GO analysis obtained 127 cellular biological processes(BP),10 cellular components(CC),20 molecular functions(MF),127 KEEG pathways,and the first 20 were selected for visualization.Conclusion Traditional Chinese medicine treatment of LDH with deficiency of liver and kidney is the core treatment method,supplemented by invigorating Qi and promoting blood circulation,eliminating arthralgia and relieving pain.The core traditional Chinese medicine treatment of LDH with liver and kidney deficiency is characterized by multiple targets,multiple pathways and multiple pathways.
