摘要:目的:观察芍药苷对葡聚糖硫酸钠(DSS)诱导慢性溃疡性结肠炎(UC)大鼠模型中炎症因子IL-17表达的影响,探讨芍药苷干预慢性溃疡性结肠炎的可能机制。方法:40只雄性Wistar大鼠随机分为4组:空白对照组、DSS组、DSS+芍药苷组、DSS+柳氮磺吡啶组。柳氮磺吡啶组作为阳性对照。除空白对照组外,其余3组大鼠采用DSS自由饮水法构建慢性溃疡性结肠炎大鼠模型。每轮DSS饮用后记录大鼠体重、粪便性状及便血情况,计算疾病活性指数(DAI);ELISA测定外周血IL-17、TNF-α及ICAM的含量;免疫组织化学法测定结肠组织IL-17蛋白表达;Western blot法测定结肠组织IL-17、LC3B及Beclin1蛋白水平。结果:与空白对照组相比,DSS组大鼠DAI评分、外周血IL-17、TNF-α及ICAM含量、结肠组织IL-17阳性区域吸光度值、结肠组织IL-17蛋白水平显著增加(P<0.01),结肠组织中LC3BⅡ及Beclin1蛋白水平显著降低(P<0.01)。与DSS组相比,DSS+芍药苷组和DSS+柳氮磺吡啶组大鼠DAI评分、外周血IL-17、TNF-α及ICAM含量、结肠组织IL-17阳性区域吸光度值、结肠组织IL-17蛋白水平下降(P<0.05),结肠组织中LC3BⅡ及Beclin1蛋白水平显著升高(P<0.05)。结论:芍药苷可能通过下调IL-17的表达,减少炎性因子IL-17、TNF-α及ICAM分泌,上调自噬相关因子LC3BⅡ及Beclin1的表达,减轻DSS对模型大鼠的影响。
Objective:To observe the effect of paeoniflorin on the expression of inflammatory factor IL-17 in rat model of chronic ulcerative colitis(UC)induced by dextran sulfate sodium(DSS),and to explore the possible mechanism of paeoniflorin in the intervention of chronic ulcerative colitis.Methods:40 male Wistar rats were randomly divided into four groups:control,DSS,DSS+paeoniflorin,DSS+sulfasalazine.Sulfasalazine was used as positive control.In addition to the control group,the rat model of chronic ulcerative colitis was established by DSS in the other three groups.Body weight,stool consistency and the presence of rectal bleeding of rats were recorded after each DSS-drinking round to calculate the disease activity index(DAI).Levels of IL-17,TNF-αand ICAM in peripheral blood were detected by ELISA.The expression of IL-17 in colon tissue was determined by immunohistochemistry.The levels of IL-17,LC3B and Beclin1 protein in colon tissues were determined by Western blot.Results:Compared with the control group,the DAI score,the levels of IL-17,TNF-αand ICAM in the peripheral blood,the absorbance value of IL-17 positive area and the IL-17 protein level in colon tissue were significantly increased in DSS group(P<0.01),while the levels of LC3BⅡand Beclin1 protein in colon tissue was significantly decreased(P<0.01)in DSS group.Compared with the DSS group,the DAI scores,the levels of IL-17,TNF-αand ICAM in the peripheral blood,the absorbance value of IL-17 positive area and the IL-17 protein level in colon tissue were markedly decreased(P<0.05),and the level of LC3BⅡand Beclin1 protein were significantly increased(P<0.05)in DSS+paeoniflorin group and DSS+sulfasalazine group.Conclusion:Paeoniflorin may weaken the effect of DSS on model rat through down-regulating the expression of IL-17,reducing the secretion of inflammatory factors IL-17,TNF-αand ICAM,and up-regulating expression of autophagy related factors LC3BⅡand Beclin1.
