摘要:目的:通过构建小鼠4T1乳腺癌细胞移植瘤Balb/c小鼠模型,探讨黄芪注射液对移植瘤Cyclin1、Bcl-2及Caspase-3蛋白表达的影响。方法:体外培养小鼠乳腺癌4T1细胞,注射接种于Balb/c小鼠右侧腹股沟皮下构建乳腺癌移植瘤模型。50只成瘤小鼠随机分为模型对照组、黄芪注射液2. 60 g/kg、5. 20 g/kg、10. 40 g/kg组和顺铂2. 5×10^-3g/kg组。观察黄芪注射液对移植瘤生长的影响;HE染色观察移植瘤形态改变;流式细胞仪分析移植瘤细胞周期;免疫组织化学法检测移植瘤组织细胞周期蛋白D1(Cyclin D1)蛋白表达;Western Blot法检测移植瘤组织Cyclin D1蛋白及凋亡相关蛋白Bcl-2、Cleaved caspase-3蛋白水平。结果:不同浓度黄芩注射液对小鼠乳腺癌移植瘤生长有显著抑制作用,黄芪注射液2. 6 g/kg、5. 2 g/kg、10. 4 g/kg抑瘤率分别为9. 1%、27. 3%、45. 5%。与模型对照组相比较,黄芪注射液5. 2 g/kg、10. 4 g/kg组及顺铂组移植瘤体积和体质量明显降低(P <0. 05或P <0. 01),G1期细胞比例明显增高,S期细胞比例显著降低(P <0. 05或P <0. 01)。黄芪注射液2. 6 g/kg、5. 2 g/kg、10. 4 g/kg组、顺铂组移植瘤组织Cyclin D1阳性区域吸光度值显著降低,Cyclin D1、Bcl-2蛋白水平明显下调,而Cleaved caspase-3蛋白水平显著上调(P <0. 05或P <0. 01)。结论:黄芪注射液对小鼠4T1乳腺癌细胞移植瘤生长有显著抑制作用,其机制与黄芪注射液下调Cyclin D1蛋白表达,促进肿瘤细胞凋亡,抑制肿瘤细胞增殖有关。
Objective: To investigate the effect of astragalus injection on the expressions of Cyclin1,bcl-2 and caspase-3 in the transplanted tumor of Balb/c mice model of 4 T1 breast cancer cell. Methods: Breast cancer 4 T1 cells were cultured in vitro and injected into the right groin of Balb/c mice to establish the breast cancer transplantation tumor model. 50 mice with transplanted tumor were randomly divided into five groups: the model group,astragalus injection groups( 2. 60 g/kg,5. 20 g/kg,10. 40 g/kg) and cisplatin group( 2. 5 × 10^-3 g/kg). The effect of astragalus injection on the growth of transplanted tumor was observed. Morphological changes of the transplanted tumor were observed by HE staining. The cell cycle of transplanted tumor was measured by flow cytometry. The expression of Cyclin D1 in transplanted tumor was detected by immunohistochemistry. The levels of Cyclin D1 protein and apoptosis-related proteins Bcl-2 and Cleaved caspase-3 in transplanted tumor were detected by Western Blot. Results: Different concentrations of astragalus injection showed significant inhibitory effect on the growth of transplanted tumor in vivo. The inhibition rates in astragalus injection groups( 2. 6 g/kg,5. 2 g/kg,10. 4 g/kg) were 9. 1 %,27.3 %,45. 5 %,respectively. Compared with the model group,the volume and mass of transplanted tumor in astragalus injection groups( 5. 2 g/kg,10. 4 g/kg) and cisplatin group were significantly reduced( P < 0. 05 or P < 0. 01). The proportion of cells in G1 stage was significantly increased,while the proportion of S stage cells was remarkable decreased( P < 0. 05 or P < 0. 01). The absorbance value of Cyclin D1 positive areas in the transplanted tumor of astragalus injection groups( 2. 6 g/kg,5. 2 g/kg,10. 4 g/kg) and cisplatin group were significantly decreased. The levels of Cyclin D1 and Bcl-2 protein were significantly down regulated,while the level of Cleaved caspase-3 protein was up regulated.( P < 0. 05 or P < 0. 01) Conclusions: Astragalus injection has a significantly inhibitory effect on the growth of transplanted 4 T1 breast cancer cells in mice. Its mechanism is related to down-regulating the expression of Cyclin D1 protein,promoting the apoptosis and inhibiting the proliferation of tumor cells.
