摘要:目的:观察消定膏对兔胫骨骨折愈合中骨钙素、转化生长因子-β1(TGF-β1)、骨形态发生蛋白-2(BMP-2)、骨形态发生蛋白-7(BMP-7)不同时相表达的影响。方法:采用骨缺损法造成家兔胫骨3mm骨缺损模型,随机分为对照组和消定膏组,外敷消定膏治疗后,分别于第7天、第14天、第28天、第42天4个不同时相点每组随机抽取8只,心脏取血,ELISA法检测血清骨钙素浓度,取骨痂组织,光镜下观察病理形态学变化,免疫组化法与RT-PCR技术检测骨痂中TGF-β1、BMP-2、BMP-7蛋白表达情况。结果:局部应用0.5g/cm^2消定膏后,与对照组比较,消定膏组血清骨钙素浓度值在各个时相点均高于对照组,消定膏组骨痂出现时间早、骨痂量多、骨折愈合时间明显缩短。消定膏组BMP-2、BMP-7、TGF-β1免疫组化MOD值、RT-PCR表达量在4个不同时相点均明显高于对照组,且在术后14天左右出现峰值,明显早于对照组(28天)。结论:消定膏对兔胫骨骨折愈合的促进作用可能通过调控骨痂组织中TGF-β/BMPs信号通路有关。
Objective :To investigate the influence of Xiaoding Ointment on expressions of TGF-β1, BMP-2 and BMP-7 in different stages during rabbits tibia fracture healing. Methods:3mm bone defect model of rabbit tibia was made by bone defect method. The rabbits were randomly divided into model group and Xiaoding Ointment group. 8 experimental rabbits were randomly selected at 4 different time points ( on the 7th day, the 14th day, the 28th day and the 42nd day). Pathomorphological changes of callus was observed under light microscope and expres- sions of TGF-β1, BMP-2 and BMP-7 in callus were detected by Immunohistochemistry and RT-PCR technique. Results:Mter topical appli- cation of Xiaoding Ointment(0. 5g/cm^2) , compared with the model group, the callus of Xiaoding group early appeared, the callus volume was much more, and the healing time was obviously shortered. The expressions of BMP-2, BMP-7 and TGF-β1 in Xiaoding Ointment group were significantly higher than those in model group at different time points ( P 〈 0.05 ), which was significantly earlier than that in model group (28 days). Conclusion:The promoting effect of Xiaoding Ointment on the healing of rabbits tihial fracture may be related to the regu- lation of TGF-β/BMPs signal pathway in callus tissue.
