摘要:目的利用两样本孟德尔随机化(MR)方法探究咖啡摄入量与骨质疏松症(OP)的因果关系。方法以咖啡摄入量为暴露因素,以OP为结局变量。咖啡摄入量和OP数据分别来源于IEU Open GWAS和GWAS Catalog数据库。显著性阈值设置为P<5×10^(-8),分析参数r^(2)=0.001,聚集窗口>10000 kb,F>10为强相关,利用MR-PRESSO进行离群值检测,筛选单核苷酸多态性(SNPs)位点作为工具变量。采用MR-Egger回归法、加权中位数(WM)法和随机效应逆方差加权(IVW)法进行MR分析,以OR值及95%CI评估咖啡摄入量与OP的因果关系。Cochran’s Q检验评估异质性,MR-Egger截距法评估水平多效性,“留一法”进行敏感性分析。结果共筛选出38个工具变量,随机效应IVW法、WM法结果显示,咖啡摄入量与OP的发生存在因果关联(P<0.05);MR-Egger回归法结果显示,咖啡摄入量与OP的发生分险不存在因果关联(P>0.05)。Cochran’s Q检验结果显示,咖啡摄入量与OP的MR分析结果不存在异质性(P>0.05);MR-Egger截距法结果接近于0,不存在水平多效性;“留一法”未发现敏感的SNPs。结论咖啡摄入量与OP的发生存在因果关联,咖啡摄入会降低OP的发生风险。
Objective To investigate the causal association between coffee intake and osteoporosis(OP)using the two-sample Mendelian randomization(MR)method.Methods Caffeine intake was used as the exposure factor,and OP was used as the outcome variable.Data on coffee intake and OP were obtained from the IEU Open GWAS and GWAS Catalog databases,respectively.The significance threshold was set at P<5×10^(-8),with analysis parameters of r^(2)=0.001,a clumping window>10000 kb,and F>10 indicating strong correlation,and MR-PRESSO was used for outlier detection to select single nucleotide polymorphisms(SNPs)as instrumental variable.MR-Egger regression method,weighted median(WM)method,and random effect inverse variance weighted(IVW)method were used for MR analysis,the causal association between coffee intake and OP was evaluated using OR value and 95%CI.Cochran’s Q test was used to evaluate heterogeneity,the MR-Egger intercept method was used to assess horizontal pleiotropy,and the“leave-one-out”method was conducted to perform sensitivity analysis.Results A total of 38 instrumental variables were selected.The results of the random effect IVW method and WM method showed that there was a causal association between coffee intake and incidence of OP(P<0.05);the result of MR-Egger regression method showed that there was no causal association between coffee intake and incidence of OP(P>0.05).Cochran’s Q test showed that there was no heterogeneity between coffee intake and MR analysis of OP(P>0.05);the result of MR-Egger intercept method was close to 0 and there was no horizontal pleiotropy;no sensitive SNPs were identified using the“leave-one-out method”.Conclusion There is a causal association between coffee intake and incidence of OP,coffee consumption reduces the risk of OP.
