摘要:目的探讨牛蒡子-甘草药对不同配伍比例(1∶0、0∶1、1∶1、1∶2、2∶1)下12个活性成分含量和抗炎作用的差异性。方法采用高效液相色谱(HPLC)法建立同时检测牛蒡子-甘草药对中12个活性成分的定量分析方法。采用二甲苯致小鼠耳肿胀、甲醛致小鼠足肿胀模型,比较不同配伍比例牛蒡子-甘草药对的抗炎作用差异。结果建立了同时检测牛蒡子-甘草药对中12个成分的HPLC色谱条件和定量方法,方法准确度高,重现性好。牛蒡子-甘草(1∶1、1∶2、2∶1)组中牛蒡子各成分溶出总量大于(1∶0)组,结果有统计学意义(P<0.05),且牛蒡子-甘草(1∶2)组中12个成分的溶出总量最大;牛蒡子-甘草(1∶2、2∶1)组中甘草各成分溶出总量大于(0∶1)组,(1∶1)组中甘草各成分溶出总量小于(0∶1)组,但结果均无统计学意义(P>0.05)。牛蒡子-甘草(1∶0、0∶1、1∶1、1∶2、2∶1)组对小鼠耳肿胀、足肿胀均有明显抑制作用,与空白对照组比较有统计学意义(P<0.05),且牛蒡子-甘草(1∶1、1∶2、2∶1)组的抑制率高于(1∶0、0∶1)组,其中以牛蒡子-甘草(1∶1)组的抑制率最高。结论牛蒡子-甘草配伍合煎后较有利于12个活性成分总量的溶出,且配伍比例为1∶2时溶出量最大;牛蒡子-甘草配伍后在抗炎作用方面产生了协同作用,其中以牛蒡子-甘草(1∶1)组的抗炎作用最佳。
Objective To study the difference of the content of 12 active components and anti-inflammatory effect of different compatibility proportions(1∶0,0∶1,1∶1,1∶2,2∶1)of Fructus Arctii-Radix Glycyrrhizae.Methods A HPLC method for simultaneous determination of 12 active components in Fructus Arctii-Radix Glycyrrhizae was established.Mice auricle swelling caused by xylene and mice foot swelling caused by formaldehyde were used to compare the anti-inflammatory effect of different compatibility proportions of Fructus Arctii-Radix Glycyrrhizae.Results The quantitative analysis method for simultaneous determination of 12 active components in Fructus Arctii-Radix Glycyrrhizae has high accuracy and good reappearance.The total disseolved amount of each component of Radix Glycyrrhizae in the Fructus Arctii-Radix Glycyrrhizae(1∶1,1∶2,2∶1)group was higher than that in the(1∶0)group(P<0.05),and the total dissolved amount of 12 components in the Fructus Arctii-Radix Glycyrrhizae(1∶2)group was the largest.When the compatibility ratio of Fructus Arctii-Radix Glycyrrhizae was(1∶2)and(2∶1),the total dissolution of components in Radix Glycyrrhizae is greater than that of its single decoction solution(0∶1)group.The total dissolution of components of Radix Glycyrrhizae in Fructus Arctii-Radix Glycyrrhizae at a ratio of(1∶1)was less than that of its single decoction solution(0∶1),but the results were not statistically significant(P>0.05).Different compatibility proportions(1∶0,0∶1,1∶1,1∶2,2∶1)of Fructus Arctii-Radix Glycyrrhizae had obvious inhibitory effects on mice ear swelling and foot swelling and showed statistically significance comparing to the blank control group(P<0.05).Moreover,the swelling inhibition rate of combined compound(1∶1,1∶2,2∶1)group was higher than that of(1∶0,0∶1)group.The swelling inhibition rate of Fructus Arctii-Radix Glycyrrhizae at a ratio of(1∶1)was the highest.Conclusion Mixed decocting of Fructus Arctii and Radix Glycyrrhizae was beneficial to the dissolution of the total amount of 12 active components,and the total dissolution of 12 components in Fructus Arctii-Radix Glycyrrhizae at a ratio of(1∶2)was the highest.The compatibility of Fructus Arctii and Radix Glycyrrhizae has synergistic anti-inflammatory effect,and the anti-inflammatory effect of Fructus Arctii-Radix Glycyrrhizae at a ratio of(1∶1)was the strongest.
