摘要:目的:观察半夏泻心汤合四神丸加减方对大鼠溃疡性结肠炎模型的影响,为临床用药的有效性提供实验依据。方法:采用异种异体结肠黏膜组织致敏法和乙酸局部灌肠相结合的方法制造动物模型。将wistar大鼠随机分为空白对照组,模型对照组,阳性对照组,半夏泻心汤合四神丸加减方颗粒剂高、中、低剂量组。连续给药14天,末次给药后24小时,处死大鼠后制备结肠组织匀浆,生化法检测结肠组织中一氧化氮(NO)、一氧化氮合酶(NOS)及TNF-α的水平。结果:半夏泻心汤合四神丸加减方可明显降低结肠组织中NO、NOS及TNF-α的水平,降低结肠病理组织学评分。结论:半夏泻心汤合四神丸加减方对溃疡性结肠炎模型大鼠有显著的干预作用,其机制可能与抑制NO生成和降低TNF-α有关。
Objective:To observe the effects of BanXia XieXinTang combining with SiShenWan on rats with ulcerative colitis and offer experimental proof for the effectiveness of clinical usage.Methods:Ulcerative colitis models were established by immune method combined with local stimulation of 5% acetic acid.Wistar rats were randomly divided into six groups.Rats in positive control group were treated with sulfasalazine,and those in three treatment groups were treated with low,middle and high dosage of BanXia XieXinTang combining with SiShenWan respectively for 14 days continuously.After 24 hours in administration,the rats were killed to obtain colon mucous membrane to test NO,NOS and TNF-α concentration of the homogenate of colon mucous membrane.Results:BanXia XieXinTang combining with SiShenWan could markedly decrease the levels of NO,NOS and TNF-α and decrease histopathological score.Conclusion:BanXia XieXinTang comhining sishenwan granules exert its marked protecting effects on rat of immune method combined with local stimulation induced ulcerative colonitis through resisting oxygen free radical lesion,inhibiting the generation of NO、NOS and TNF-α.
