摘要:目的考察聚酰胺树脂分离纯化藏药湿生扁蕾总黄酮的最佳工艺。方法 70%乙醇回流提取湿生扁蕾,提取液回收乙醇后上聚酰胺树脂柱吸附,用不同体积分数乙醇洗脱,比色法检测洗脱液总黄酮的含量。结果聚酰胺树脂能很好地吸附湿生扁蕾中的总黄酮,70%乙醇洗脱总黄酮的回收率为90.32%。聚酰胺树脂在适当处理后可重复利用,但随着重复利用次数的增加,聚酰胺树脂对黄酮的吸附力不断下降。结论湿生扁蕾提取物经聚酰胺树脂分离纯化能得到较纯的总黄酮,且工艺简便易行,得率高,为工业化生产提供了参考依据。
Objective To investigate the optimum process on separation and purification of total flavonoid in Gentianopsis paludosa ( Mum. ) Ma by polyamide resin. Methods Gentianopsis paludosa ( Mum. ) Ma was extracted by circulation reflux of 70% ethanol, the extracting solution was adsorbed on polyamide resin column after the etha- nol was recovered, then eluted with different concentrations of ethanol, detected the content of total flavonoid in elu- tions by colorimetric method. Results Polyamide resin could adsorb the total flavonoid in Gentianopsis paludosa ( Mum. ) Ma very well, the retrieve ratio of the total flavonoid was 90.32% by 70% ethanol, the content of the elu- tion was 71.21%. Polyamide resin could be utilized repeatedly after disposed properly, but the capacity of adsorp- tion was declining with the times was increasing. Conclusion dosa( Mum. )Ma can be found by separation and purification of can offer reference for industrialization production.
