摘要:目的:探讨菖蒲郁金汤对多发性抽动症模型大鼠行为及脑组织IL-6及TNF-α的影响。方法:82只雄性大鼠随机分为空白组12只,造模组70只,采用亚氨基二丙腈(IDPN)腹腔注射法制备多发性抽动症动物模型,造模成功后,除去体重偏差较大的10只大鼠,将造模组其余60只大鼠再次分为:模型组、泰必利组、菖蒲郁金汤高、中、低剂量组,各12只。从造模成功后第2天开始给予灌胃,泰必利组给予泰必利(47.91mg/kg),菖蒲郁金汤各剂量组给予菖蒲郁金汤(77.28g/kg、38.64g/kg、19.32g/kg),空白组与模型组给予等体积纯净水,连续给药4周。造模成功后及每周灌胃后1小时,观察大鼠行为。在最后一次给药后24小时计算胸腺指数、脾脏指数,用ELISA法检测IL-6、TNF-α水平;观察纹状体组织病理学变化。结果:1造模后,各组大鼠均出现刻板行为和运动行为;治疗4周后,泰必利组及菖蒲郁金汤组(77.28/kg、38.64g/kg)刻板行为评分明显下降;泰必利组及菖蒲郁金汤组(77.28/kg)运动行为评分明显下降。2模型组大鼠胸腺指数和脾脏指数均明显下降;菖蒲郁金汤高剂量(77.28g/kg)组可明显提高胸腺指数和脾脏指数。3各组大鼠脑组织中IL-6含量无明显变化;模型组大鼠脑组织中TNF-α水平明显下降;泰必利(47.91mg/kg)组大鼠脑组织中TNF-α水平无明显变化;菖蒲郁金汤高剂量(77.28/kg)组大鼠脑组织中TNF-α水平明显升高。4空白组大鼠纹状体神经细胞分布均匀、大小适中、形态如常;模型组大鼠纹状体神经细胞减少,以神经元细胞减少为主,且神经元细胞体积缩小,形态不规则;泰必利(47.91mg/kg)组大鼠纹状体神经细胞数量及形态变化与模型组无明显区别;菖蒲郁金汤各剂量(77.28g/kg、38.64g/kg、19.32g/kg)组纹状体神经元细胞数量及形态均有所改善,以高剂量(77.28g/kg)组为最显著。结论:菖蒲郁金汤具有抗抽动作用,其抗抽动作用可能与改善多发性抽动症模型大鼠免疫功能及修复神经细胞有关。
Objective: To observe the effects of Changpu Yujin( CY) decoction on the behavior and IL-6,TNF-α in brain tissue of tourette syndrome( TS) in rat model.Methods: The 82 male SD rats were randomly divided into 12 in blank group and 70 in control group,the control group was built up by intraperitoneal injecting IDPN.Then 10 rats in control group were removed from those because their weights were beyond the standard range.60 rats in control group were randomly divided into model group,tiapride group,high dose of CY decoction group,middle dose of CY decoction group and low dose of CY decoction group again,12 in each group.The blank group and the model group were intragastricly perfused by pure water,and the remaining four groups were given tiapride( 47.91mg/kg) and CY decoction( 77.28g/kg,38.64g/kg,19.32g/kg) respectively,4 consecutive weeks.The rat behaviors were observed after the modeling and after administration 1 hour weekly.Thymus index and spleen index were calculated; The levels of IL-6,TNF-α were measured by ELISA; The histopathological changes in the striatum were observated 24 hours after the last administration.Results: 1After modeled,the stereotypical behavior and motor behavior were observed in all groups,and there was no significant difference between the two groups( P 〉 0.05).Four weeks after treatment,the scores of the stereotyped behavior of rats in the tiapride group and CY decoction group( 77.28/kg,38.64g/kg) significantly decreased,and compared with the model group,there was significant difference in them( P 〈 0.01); the scores of the motor behavior of rats in the tiapride group and CY decoction group( 77.28/kg) significantly decreased,and compared with the model group,there was difference in them( P 〈 0.05).2Compared with the blank group,the thymus index and spleen index decreased significantly in the model group( P 〈 0.01,P 〈 0.01).Compared with the model group,the thymus index and the spleen index increased significantly in the high dose of CY decoction( 77.28g/kg)group( P 〈 0.01,P 〈 0.05).3The level of IL-6 of rats in each group had no significant change( P 〉 0.05); Compared with the blank group,the level of TNF-α decreased significantly in the model group( P 〈 0.01).Compared with the model group,the level of TNF-α was unchanged obviously in the tiapride( 47.91mg/kg) group( P 〉 0.05),the level of TNF-α was increased significantly in the high dose of CY decoction( 77.28/kg) group( P 〈 0.01).4The nerve cells in the striatum of the blank group were distributed evenly,its size was medium,and its shape was usual; The nerve cells in the striatum of the model group were decreased; The reduction of neuronals was the main,and the volume of neuronals was narrow,its shape was irregular; Compared with the model group,the number and shape of the nerve cells in the striatum of the tiapride( 47.91mg/kg) group had no significant change; The number and morphology of the nerve cells in the striatum of the high/middle/low dose of CY decoction( 77.28g/kg,38.64g/kg,19.32g/kg) group were improved,and the improve of the high dose( 77.28g/kg) group was the most significant.Conclusion: The CY decoction has anti-tic effects,its anti-tic ? effects may be related with improving immune function and repairing of nerve cells in a rat model of TS.
