摘要:目的观察黄芪多糖(APS)对KK-Ay小鼠肾组织血管内皮损伤的影响。方法将C57BL/6J小鼠设为空白组,将KK-Ay小鼠通过喂养高脂高糖饲料建立糖尿病肾病模型,造模后随机分为模型组、对照组(25 mg·kg^(-1)厄贝沙坦灌胃)和低、中、高实验剂量组(100、200、400 mg·kg^(-1)APS灌胃),每日灌胃1次,连续4周。给药干预4周后,心脏取血。用免疫组织化学染色方法测定小鼠肾组织中内皮素-1(ET-1)、血管性血友病因子(vWF)的蛋白表达水平;用蛋白质印迹法分析小鼠肾组织中NT蛋白的表达水平;用实时荧光定量聚合酶链反应法检测小鼠肾组织中SUR亚基2B/内整流钾离子通道亚单位6.1(SUR2B/Kir6.1)mRNA的表达水平。结果空白组、模型组、对照组和低、中、高剂量实验组小鼠的ET-1蛋白表达水平分别为0.25±0.05、0.43±0.02、0.28±0.06、0.39±0.01、0.34±0.03和0.30±0.02;vWF蛋白表达水平分别为0.24±0.04、0.35±0.02、0.26±0.03、0.34±0.03、0.30±0.03和0.28±0.02;NT蛋白表达水平分别为1.00±0.03、1.67±0.06、1.05±0.05、1.39±0.08、1.37±0.07和1.14±0.05;SUR2B mRNA表达水平分别为1.00±0.02、0.18±0.01、0.83±0.02、0.29±0.01、0.46±0.02和0.63±0.02;Kir6.1 mRNA表达水平分别为1.00±0.03、0.13±0.02、0.74±0.02、0.21±0.02、0.33±0.05和0.51±0.02。对照组、高剂量实验组上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论黄芪多糖能够改善DN小鼠肾组织血管内皮损伤,保护微血管结构与功能,延缓DN病情进展。
Objective To observe the effect of astragalus polysaccharide(APS)on vascular endothelial injury in kidney tissue of KK-Ay mice.Methods C57BL/6J mice were set as blank group,KK-Ay mice were induced to become models by feeding high-fat and high-sugar chow,and KK-Ay mice were divided into the model group,the control group(irbesartan 25 mg·kg^(-1) was given by gavage),and the experimental-L,-M,-H groups(APS 100,200 and 400 mg·kg^(-1) was given by gavage),gavaged once daily for 4 weeks.Blood was taken from the heart 4 weeks after the administration of the drug.The expression levels of endothelin-1(ET-1)and vascular hemophilic factor(vWF)in renal tissues of mice were determined by immunohistochemical staining;the exp re ssion levels of NT proteins in renal tissues were analyzed by immunoprotein blotting;the expression levels of sulfonylurea receptor 2B/inward rectifyimg potassium channel 6.1(SUR2B/Kir6.1)mRNA were detected in renal tissues of mice by real-time fluorescence quantitative polymerase chain reaction.Results The expression levels of ET-1 protein in mice in the blank group,model group,control group and experimental-L,-M,-H groups were 0.25±0.05,0.43±0.02,0.28±0.06,0.39±0.01,0.34±0.03 and 0.30±0.02,respectively;the expression levels of vWF protein were 0.24±0.04,0.35±0.02,0.26±0.03,0.34±0.03,0.30±0.03 and 0.28±0.02,respectively;NT protein expression levels were 1.00±0.03,1.67±0.06,1.05±0.05,1.39±0.08,1.37±0.07 and 1.14±0.05,respectively;SUR2B mRNA expression levels were 1.00±0.02,0.18±0.01,0.83±0.02,0.29±0.01,0.46±0.02 and 0.63±0.02,respectively;Kir6.1 mRNA expression levels were 1.00±0.03,0.13±0.02,0.74±0.02,0.21±0.02,0.33±0.05 and 0.51±0.02,respectively.Compared with the model group,the differences of the above indexes in the control group and the experimental-H group were statistically significant(P<0.05,P<0.01).Conclusion Astragali polysaccharide can improve the vascular endothelial damage of renal tissues in DN mice,protect the microvascular structure and function,and delay the progression of DN disease.
