摘要:目的:比较甘草内生菌与宿主甘草对痰浊阻肺模型大鼠的祛痰作用,筛选甘草内生菌有效菌株。方法:将Wistar大鼠70只随机分为7组,即正常对照组、模型组、复方贝母氯化铵阳性对照组、野生甘草水煎液组及甘草内生菌各组(JTYF023、JTYB006、JTYB029)。采用SO2烟薰、低温及冷风刺激诱导痰浊阻肺大鼠模型。造模10 d后,给予各组大鼠相应药物灌胃,连续7 d。进行证候学观察,检测酚红排泌量、肺功能指标4项、炎症相关因子(TNF-α、ICAM-1、COX-2)的含量、肺组织Na^+-K^+-ATP酶活性,并以免疫组化法测定水通道蛋白AQP1、AQP5的分布及表达,探讨药物的祛痰机制。结果:与正常对照组比较,模型组大鼠酚红排泌量增加,肺功能4项指标显著降低,炎症各相关因子的含量显著升高,Na^+-K^+-ATP酶活性及水通道蛋白AQP1、AQP5表达显著降低(P<0.01)。与模型组比较,水煎液组、JTYB029组酚红排泌量显著降低,肺功能4项指标显著升高,炎症各相关因子的含量显著降低,Na^+-K^+-ATP酶活性及水通道蛋白AQP1、AQP5表达显著升高(P<0.05或P<0.01)。结论:筛选出1株(JTYB029)与野生甘草水煎液祛痰作用相当的内生菌。
Objective:To screen effective strains of endophytes from Glycyrrhiza uralensis by comparing expectorant effects of endo- phytes and Glycyrrhiza uralensis. Methods :70 Wistar rats were randomly divided into normal control group, model group,compound fritil- laria and ammonium chloride (Fufang Beimu Lvhuaan)group, wild Glycyrrhiza uralensis water decoction group, three dose endophytes groups( JTYF023 ,JTYB006 ,JTYB029 ), with ten rats in each group. The model of phlegm blocking in lung was induced by inhaleing SO2 and cold wind. After modeling for 10 d, all drug intervention groups were orally administrated corresponding drugs for 7 d. The gen- eral activities,the amount of phlegm and pulmonary function were measured. The pulmonary funtion, the levels of tumor necrosis factor ( TNF-a), intercellular adhesion molecule 1 ( ICAM-1 ), cyclooxygenase-2 ( COX-2), and the activity of Na ~ -K ~ -ATPase in lung tissue were measured by ELISA method. The Aquaporin-1 ( AQP1 ) and Aquaporin-5 ( AQP5 ) expression in lung tissue of rats were measured by immunohistochemical method. Results:Compared with the control group, the amount of phlegm significantly increased and pulmonary function were significantly decreased in model group. The levels of TNF-a, ICAM-1, and COX-2 in lung tissue were significantly in- creased,the pulmonary function and the activity of Na+ -K+ -ATPase and the expression of AQP1 and AQP5 were significantly decreased (P 〈0. 01 ). Compared with the model group, in the wild Glycyrrhiza uralensis decoction group and JTYB029 group, the amount of phlegm were significantly decreased and pulmonary function were significantly increased, the levels of TNF-a, ICAM-1 and COX-2 in lung tissue significantly were decreased, the activity of Na + -K ~ -ATPase and the expression of AQP1 and AQP5 were significantly in- creased(P 〈0. 01 or P 〈0. 05). Conclusion:The strains of Endophytes(JTYB029)and wild Glycyrrhiza uralensis water decoction have expectorant effect.
