摘要:目的:探讨不同干燥工艺对红芪黄酮类成分毛蕊异黄酮和芒柄花素含量的影响,为其最佳干燥工艺的建立提供实验依据。方法:红芪经不同方法干燥后,采用HPLC测定红芪中的毛蕊异黄酮和芒柄花素含量并比较;采用Agilent1260高效液相仪液C18柱;乙腈-0.01%磷酸水溶液为流动相;洗脱梯度,0~12min,30%~33%乙腈;12~13min,33%~40%乙腈,13~25min,40%乙腈;流速为1.0mL·min^-1;选用248nm为检测波长;色谱柱温度为30℃;进样量为10μL。结果:红芪电热鼓风干燥(条件:65℃,2层)组毛蕊异黄酮、芒柄花素含量最高,真空干燥组、微波干燥(条件:高温,3层)组、阴干组、真空冷冻干燥组次之,晒干组最低。结论:不同方法干燥红芪对红芪中毛蕊异黄酮和芒柄花素含量有一定的影响;电热鼓风干燥红芪效率高、方法简单、时间短、成本低,适合药材大批量干燥,可作为红芪干燥的优先选择方法。
Objective:The effects of different drying techniques on the content of calycosin and formononetin in Hedysari Radix were compared to provide the basis for establishing the best drying technology of Hedysari Radix. Methods:The contents of calycosin and formononetin in Hedysari Radix were determined by HPLC after drying by different methods.The chromatographic conditions were as follows:Agilent HC-C 18 (250 mm ×4.6 mm,5 μm );mobile phase:acetonitrile-0.01% phosphoric acid solution.The gradient elution was 0-12 min,the acetonitrile was 30%-33%;12-13 min,the acetonitrile was 33%-40%;13-25 min,the acetonitrile was 40%.The mobile phase flow rate was 1.0 ml · min^-1 ,and the detection wavelength of DAD detector was 248 nm.The column temperature was 30 ℃.The injection amount was 10 μL. Results:The content of calycosin and formononetin was the highest in Hedysari Radix dring with electrothermal blast (condition:65 ℃, 2 layers ),followed by vacuum drying group,microwave drying group (high temperature,3 layers ),drying in shade group,vacuum freeze drying group,and the lowest was sun drying group. Conclusion:Different methods of drying Hedysari Radix have certain influence on the contents of calycosin and formononetin in Hedysari Radix,the electrothermal blow-drying method has the advantages of high efficiency,simple method,short time and low cost,so it is suitable for mass drying of medicinal materials.It can be used as a preferred method for drying of Hedysari Radix.
