摘要:目的优化复合酶提取肉苁蓉多糖的最佳工艺,研究酶解后肉苁蓉多糖的传质过程。方法以酶用量、酶解温度、酶解时间为考察因素,肉苁蓉多糖提取率为考察指标,通过正交试验确定复合酶提取肉苁蓉多糖的最佳工艺,对最佳工艺条件下多糖得率随时间的动力学变化用Power law模型和Parabolic diffusion模型进行拟合。结果复合酶提取肉苁蓉多糖的最佳工艺条件为复合酶用量0.2%,酶解温度50℃,酶解时间1.5 h;肉苁蓉经复合酶辅助提取后,多糖提取率和扩散速率常数明显提高,提取特征常数、多糖扩散指数明显不同。结论酶技术作为中药提取的一种新方法,应用于濒危药材肉苁蓉多糖的提取,具有较好的前景。
Objective To optimize the enzymatic extraction process for Cistanche deserticola polysaccharide (CDPS) and study the mass transfer process of CDPS after enzymolysis. Methods Enzyme dosage, hydrolysis temperature and enzyme hydrolysis time were taken as reference factors, extraction rate of polysaccharide was used as indicator and the orthogonal experiment was adopted for the optimization. Under this optimum condition, kinetic change of polysaceharide yield with time-variation was fitting by the models of Power law and Parabolic diffusion. Results The optimum conditions were enzyme dosage 0.2% , enzymolysis temperature 50 ~C and hydrolysis time 1.5 h ; extraction rate and diffusion rate constant increased obviously, there was a significant difference among the indexes. Conclusion Enzyme technology can be used to extract CDPS as a new method for Chinese medicine, which has a good application prospect in the extraction of endangered crude drug Cistanche deserticola.
