摘要:目的通过网络药理学探讨石菖蒲-郁金药对治疗儿童抽动障碍(TD)的有效成分、作用靶点、信号通路以及可能的作用机制。方法采用中药系统药理学分析平台(TCMSP)数据库筛选石菖蒲-郁金的有效成分及相关作用靶点,使用人类基因组注释(Gene Cards)数据库得到已知治疗的儿童TD靶点,用Venny 2.1构建绘制药物-疾病靶点的韦恩图;使用蛋白-蛋白相互作用网络(STRING)数据库和Cytoscape 3.7.1软件构建关键靶点基因网络的相互作用;借助注释、可视化和集成发现(DAVID)数据库对关键靶点进行基因本体(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析;通过Cytoscape 3.7.1软件构建该组药对治疗儿童TD的有效成分-关键靶点-信号通路网络。结果筛选得到19个石菖蒲-郁金的有效成分,预测出该药对治疗儿童TD的关键靶点32个,主要与μ阿片受体1(OPRM1)、β2肾上腺素受体(ADRB2)、毒蕈碱型乙酰胆碱受体M2(CHRM2)有关,主要通过神经活性配体-受体相互作用信号转导通路等信号通路发挥治疗作用。结论石菖蒲-郁金药对组合中的多种有效成分通过多条信号通路作用于不同生物靶点,达到抗抽动的作用效果,分析成分、靶点和通路三者之间的关系可为进一步实验研究提供理论基础,有助于探明石菖蒲-郁金药对治疗儿童TD的作用机制。
Objective To explore the active components,targets,signaling pathways and possible action me-chanism of couplet medicinals of Acori Tatarinowii rhizoma and Curcumae radix in the treatment of children with tic disorders(TD)based on network pharmacology.Methods The active components and related targets of Acori Tatarinowii rhizoma-Curcumae radix were screened out by searching the Database platform of Traditional Chinese Medicine Systems Pharmacology(TCMSP).Gene Cards database was used to obtain TD targets of known treatment in children,and Venny 2.1 was used to construct a Venn diagram of drug-disease targets.Search Tool for the Retrieval of Interacting Proteins(STRING)database and Cytoscape 3.7.1 software were used to construct the interaction of key target gene network.Gene Ontology(GO)function enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)signal pathway analysis of key targets were performed by DAVID database.Cytoscape 3.7.1 software was used to construct the active component-key target-signal pathway network of this couplet medicinals for the treatment of TD.Results Nineteen active components were screened out from couplet medicinals of Acori Tatarinowii rhizoma and Curcumae radix,and 32 key targets were predicted for the treatment of TD in children,mainly withμ1-opioid receptor(OPRM1),β2 adrenoceptor(ADRB2)and muscarinic acetylcholine receptor M2(CHRM2),which play a therapeutic role mainly through the signal transduction pathway of neuroactive ligand-receptor interaction.Conclusion A variety of active components in couplet medicinals of Acori Tatarinowii rhizoma and Curcumae radix act on different biological targets through multiple signaling pathways to achieve the effect of anti twitch.The analysis of the relationship among components,targets and pathways can provide a theoretical basis for further experimental research and help to explore the action mechanism of couplet medicinals of Acori Tatarinowii rhizoma and Curcumae radix in the treatment of children with TD.
