摘要:目的:探讨不同浓度的黄芩苷对胃癌SGC-7901细胞增殖的抑制作用,以及对pten和survivin基因的影响。方法:以不同浓度(10、20、40、80、160、320μmol/L)的黄芩苷处理胃癌SGC-7901细胞后,采用MTT法测定细胞活力;80、120、160μmol/L的黄芩苷处理胃癌SGC-7901细胞后,采用RT-q PCR法检测Toll样受体8(TLR8)、低氧诱导因子(HIF-1α)、血小板衍生生长因子β(PDGF-β)和第10号染色体上磷酸酶和张力白同源缺失性基因(pten)的表达,Western-blot法分别检测胃癌细胞TLR8、HIF-1α、PDGF-β和pten蛋白的表达。结果:不同浓度的黄芩苷均能抑制细胞增殖,RT-q PCR法检测显示TLR8、HIF-1α、PDGF-β基因表达下调,pten基因表达上调。结论:不同浓度的黄芩苷可抑制胃癌细胞增殖,并能上调pten和下调TLR8、HIF-1α、PDGF-βm RNA及蛋白的表达。
Objective: To discuss the inhibiting effects of baicalin with different concentrations on the proliferation of gastric cancer SGC-7901 cell, and its influence on the gene pten and survivin. Methods: Cellular activity was measured by adopting MTT method after dealing SGC-7901 cell with baiealin with different concentrations (10, 20, 40, 80, 160 and 320 μmol/L); the expressions of TLR8, HIF-1α, PDGFβ and pten were detected by RT-qPCR method after dealing SGC-7901 cell with baicalin with different concentrations (80, 120 and 160 μmol/L); the expressions of TLR8, HIF-1α , PDGFβ and pten were detected by Western-blot method respectively. Results: Baicalin with different concentrations could inhibit cellular proliferation, RT-qPCR method showed that the expressions of TLR8, HIF-1α and PDGFβ down regulated, while pten expression up regulated. Conclusion: Baicalin with different concentrations could inhibit gastric cancer cellular proliferation, and it could up regulate pten expressions and down regulate the expressions of TLR8, HIF-1α and PDGFβ mRNA and protein.
