摘要:目的:探讨参苓白术散对脾虚湿困型溃疡性结肠炎(UC)的治疗作用及其对MKK/JNK通路的影响。方法:采用综合法复建脾虚湿困型UC大鼠模型,给予参苓白术散干预治疗。观察大鼠一般状况;计算大鼠免疫器官指数;对结肠组织进行病理学检测;ELISA法检测血清及结肠中IL-2、IL-17含量;RT-PCR测定结肠MKK4、MKK7、JNK、AP-1 mRNA表达水平;Western blot检测结肠MKK4、MKK7、JNK、AP-1蛋白及其磷酸化蛋白表达水平。结果:与空白组相比,UC大鼠胸腺指数显著降低,脾脏指数明显升高,血清与结肠中IL-2、IL-17含量、结肠MKK4、MKK7、JNK、AP-1 mRNA表达水平、结肠p-MKK4/MKK4、p-MKK7/MKK7、p-JNK/JNK、p-AP-1/AP-1值均显著升高;与模型组相比,干预组胸腺指数均有所上升,脾脏指数均有所下降,其余上述指标均有不同程度的降低,以参苓白术散高剂量组降低最明显。结论:参苓白术散可减轻脾虚湿困型UC大鼠结肠组织炎症反应,其作用机制可能是通过抑制MKK/JNK信号通路激活从而减轻炎症。
Objective:To explore therapeutic effect of Shenlingbaizhu Powder in treating ulcerative colitis(UC)of spleen deficiency and dampness type and its influence on MKK/JNK pathway.Methods:UC rats model with spleen deficiency and dampness were rebuild by a comprehensive method,and Shenlingbaizhu Powder was given for interventional treatment.Observed general conditions of rats;calculated index of rat immune organs;conducted pathological examination of colon tissue;levels of IL-2 and IL-17 in serum and colon were measured by ELISA;colon MKK4,MKK7,JNK,AP-1 mRNA expression levels were measured by RT-PCR;Western blot was used to measure expression levels of colonic MKK4,MKK7,JNK,AP-1 proteins and their phosphorylated proteins.Results:Compared with blank group,thymus index of UC rats was decreased significantly,spleen index was increased significantly,levels of IL-2 and IL-17 in serum and colon,mRNA expression levels of colonic MKK4,MKK7,JNK,AP-1,and colonic p-MKK4/MKK4,p-MKK7/MKK7,p-JNK/JNK,p-AP-1/AP-1 values were all increased significantly;compared with model group,thymus indexes of intervention groups were increased,and spleen indexes were increased,and the rest of the above indicators were all decreased to varying degrees,and the decrease was more obvious in high-dose group of Shenlingbaizhu Powder.Conclusion:Shenlingbaizhu Powder can alleviate inflammation in colon tissue of UC rats with spleen deficiency and dampness.Its mechanism may be through inhibiting the activation of MKK/JNK signaling pathway,thereby reducing inflammation.
