摘要:目的:观察生骨再造丸对激素性股骨头坏死(SANFH)大鼠炎症因子IL-4、IL-6及TNF-αmRNA及蛋白表达的影响,以进一步评价生骨再造丸对激素性股骨头坏死的治疗作用机制。方法:60只6周龄雄性SD大鼠随机分为正常对照组、激素模型组、生骨再造丸0. 5 g/kg、1 g/kg、2 g/kg组,健骨生丸1. 68 g/kg组,每组各10只。除正常对照组外,其余5组采用内毒素加激素联合造模,造模第6周末每组随机处死1只大鼠,检验造模是否成功。第7周开始,各组灌胃相应药物或生理盐水,连续给药8周。眼球取血分离血清,ELIAS法检测三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)水平,血清钙(SCa)、血清磷(S-P)、血清骨保护素(OPG)、骨钙素(OCN)、血清碱性磷酸酶(ALP)、核因子-κB受体活化因子配体(RANKL)、Ⅰ型前胶原羧基末端前肽(PINP)水平活性;采用RT-PCR和Western blot法观察骨组织中白介素-4(IL-4)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α) mRNA和蛋白表达情况。结果:与正常组比较,模型组血清TG、TC、LDL及RANKL、ALP、OPG含量明显升高,HDL、SCa、S-P、OCN、PINP含量明显下降;IL-4、IL-6、TNF-αmRNA及蛋白表达水平均明显上调。与模型组比较,各治疗组TG、TC、LDL及OPG、RANKL、ALP含量明显降低,HDL、S-Ca、S-P、OCN及PINP含量明显升高;IL-4、IL-6、TNF-αmRNA及蛋白表达水平均明显下调。结论:生骨再造丸对激素性股骨头坏死大鼠血脂及骨代谢相关指标有明显调节作用,可有效抑制IL-4、IL-6、TNF-αmRNA及蛋白表达水平,这可能是其治疗激素性股骨头坏死的机制之一。
Objective: To observe the effect of Shengguzaizao Pill on the mRNA and protein expressions of inflammatory factors IL-4,IL-6 and TNF-alpha of rats with steroid-induced avascular necrosis of femoral head (SANFH),and to further evaluate its therapeutic mechanism.Methods: 60 six-week-old male SD rats were randomly divided into the control group,hormone model group,Shengguzaizao pill groups (0.5 g/kg、1 g/kg、2 g/kg) and 1. 68 g/kg Jiangusheng group,10 rats in each group. Except for the control group,endotoxin combined with hormone was used in other five groups to establish the model. At the end of the 6 th week,one rat in each group was randomly taken to be sacrificed to check whether the model successfully established. From the 7 th week,corresponding drugs and saline water were administered to rats,for 8 weeks. After administration,blood samples were collected from eyeballs and serum levels of triglyceride (TG),total cholesterol (TC),high density lipoprotein (HDL),low density lipoprotein (LDL),S-Ca,S-P,serum osteoprotegerin (OPG),osteocalcin (OCN),alkaline phosphatase (ALP),nuclear factor-kB receptor activator ligand (RANKL) and type I procollagen carboxyl terminal pro-peptide (PINP)were measured by ELISA. The mRNA and protein expressions of interleukin-4 (IL-4),interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) in bone tissues were observed by RT PCR and western bolt. Results: Compared with the control group,serum contents of TG,TC,LDL,RANKL,ALP and OPG in the model group were significantly increased,contents of HDL,OPG,S-Ca,S-P,OCN and PINP were significantly decreased,mRNA and protein expressions of IL-4,IL-6 and TNF-alpha were significantly up regulated. Compared with the model group,contents of TG,TC,LDL,OPG,RANKL and ALP were significantly decreased,contents of HDL、S-Ca、S-P、OCN and PINP were significantly increased,mRNA and protein expression levels of IL-4,IL-6,TNF-alpha were significantly down regulated. Conclusion:Shengguzaizao Pills can significantly regulate the serum lipid and bone metabolism of rats with steroid-induced femoral head necrosis,it can effectively inhibit the mRNA and protein expressions of IL-4,IL-6 and TNF-alpha,which may be one of the mechanisms on treating steroid-induced femoral head necrosis.
