摘要:目的:通过观察野菊花提取物对自发性高血压大鼠(SHR)血管紧张素转换酶2(ACE_(2))表达及氧化应激水平的影响,探讨其作用机制。方法:将50只SHR大鼠随机分为5组,即模型对照组、贝那普利0.9 mg/kg组、野菊花提取物1.4、2.8、5.6 g/kg组,另设一正常对照组,各组连续灌胃生理盐水或相应的药物28 d。测量给药前后大鼠的血压,并于给药结束后,检测主动脉组织和血清中SOD、GSH-Px、MDA活性,采用PCR法和WB法检测主动脉组织中ACE_(2)的表达。结果:与正常对照组比较,模型对照组收缩压、舒张压明显升高,ACE_(2)表达减弱,SOD、GSH-Px活性降低、MDA含量升高(P<0.05或P<0.01);与模型对照组比较,野菊花提取物组和贝那普利0.9 mg/kg组收缩压和舒张压降低,ACE_(2)表达增强,SOD、GSH-Px活性增强,MDA含量降低(P<0.05或P<0.01)。结论:野菊花可能通过促进ACE_(2)表达及影响机体氧化应激水平从而产生降血压的作用。
Objective:To observe the effect of Chrysanthemi Indici Flos extract on the expression of ACE_(2)and oxidative stress level in spontaneously hypertensive rats(SHR),and to explore its mechanism.Methods:Fifty SHR rats were randomly divided into 5 groups:the model group,0.9 mg/kg benazepril group,1.4,2.8 and 5.6 g/kg Chrysanthemi Indici Flosgroups,and the control group.The rats in each group were given saline or corresponding drugs for 28 days.The blood pressures of rats were measured before and after the administration.The activities of SOD,GSH-Px,and the level of MDA in the Aortic tissue and serum were detected,and the expression of ACE_(2)in the aortic tissue was detected by PCR and WB methods.Results:Compared with the control group,the systolic blood pressure and diastolic blood pressure were significantly increased in the model group,the expression of ACE_(2)was decreased,the activities of SOD,GSH-PX were decreased,and the content of MDA was increased(P<0.05,P<0.01).Compared with the model group,the systolic blood pressure and diastolic blood pressure were decreased in 0.9 mg/kg benazepril group and Chrysanthemi Indici Flos groups,the expression of ACE_(2)was increased,the activities of SOD,GSH-PX were increased,and the content of MDA was decreased(P<0.05,P<0.01).Conclusion:Chrysanthemi Indici Flos may reduce the blood pressure by promoting ACE_(2)expression and affecting oxidative stress level.
