摘要:目的:观察黄芪及其提取物对矽肺模型大鼠支气管肺泡灌洗液(bronchial alveolus fluid,BALF)MMP-2和MMP-9表达的影响,探讨黄芪抑制矽肺纤维化的作用机制。方法:SPF级Wistar雄性大鼠120只,随机分为正常组、模型组、乙酰半胱氨酸组、黄芪水煎液组、黄芪总苷组、黄芪多糖组,每组20只。大鼠水合氯醛麻醉后,采用经气管染Si O2粉尘法复制矽肺模型。干预组注入Si O2粉尘后1d开始用黄芪提取物及黄芪总苷灌胃,正常组及模型组给予等体积的生理盐水灌胃。观察各组肺组织病理改变,行支气管肺泡灌洗术,用ELISA法检测支气管肺泡灌洗液MMP-2及MMP-9的表达。结果:各时段模型组MMP-2和MMP-9表达较正常组均降低(P<0.05),而乙酰半胱胺酸组、黄芪各干预组各时段表达均低于模型组,在28天MMP-2和MMP-9表达均下降(P<0.05)。结论:黄芪能降低矽肺模型大鼠支气管肺泡灌洗液MMP-2、MMP-9的表达,这可能是其抑制矽肺纤维化形成的机制之一。
Objective: To explore the mechanism of HuangQi(radix astragali) inhibiting silicosis by observing the influence of HuangQi and its extractive on the expressions of MMP-2 and MMP-9 in bronchial alveolus fluid(BALF) of rat model with silicosis. Methods: Altogether 120 SPF rats were randomized into the normal group, the model group, acetylcysteine group, HuangQi decoction group, total glycosides(TG) group of HuangQi and astragalus polysaccharides(APS) group, 20 rats each group. Rat model with silicosis were duplicated by SiO2 dust dyeing method through air tube after the rats accepted anaesthesia with chloral hydrate. The intervention group accepted intragastric administration of HuangQi extractive and HuangQi TG in one day after injecting SiO2 dust, the normal group and the model group accepted intragastric administration of physiological saline in the same volumes. Pathological changes of pulmonary tissue were observed in different groups and bronchoalveolar lavage was performed,the expressions of MMP-2 and MMP-9 in BALF were detected by ELISA method. Results: The expressions of MMP-2 and MMP-9 of the model group were lower compared with the normal group in different times(P〈0.05),while the expressions were lower in acetylcysteine group and HuangQi intervention groups in different times, the expressions of MMP-2 and MMP-9 were decreased at 28 days(P〈0.05). Conclusion: HuangQi could decrease the expressions of MMP-2 and MMP-9 in BALF of rat model with silicosis, and it might be one of the mechanisms of inhibiting silicosis.
