摘要:目的:比较四倍体黄芩新品系D20和甘肃地产二倍体黄芩化学成分的差异。方法:采用超高效液相色谱法建立四倍体黄芩新品系D20和甘肃地产二倍体黄芩药材的色谱指纹图谱,并对两者中9种黄酮类成分进行含量测定。结果:四倍体新品系D20和二倍体化学成分指纹图谱相似度大于0.99,二倍体黄芩中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、野黄芩苷、木犀草素、芹菜素、芹菜素-7-O-β-D-葡萄糖醛酸苷、千层纸素A质量分数分别为(14.72±0.46)%、(5.84±0.31)%、(1.55±0.49)%、(0.72±0.21)%、(0.81±0.04)%、(1.47±0.17)%、(0.55±0.01)%、(0.30±0.04)%、(0.12±0.01)%。四倍体黄芩中相应成分的质量分数分别为(16.54±1.45)%、(6.62±0.39)%、(0.28±0.01)%、(0.20±0.02)%、(0.87±0.06)%、(1.50±0.19)%、(0.19±0.04)%、(0.31±0.01)%、(0.10±0.04)%。结论:四倍体黄芩新品系D20和二倍体黄芩化学成分基本一致,但各成分含量存在一定差异,除芹菜素、黄芩素、汉黄芩素、千层纸素A的含量四倍体比二倍体低外,其他5种成分的含量四倍体均比二倍体高。
Objective:To compare the chemical constituents between the tetraploid new strains D20 and the cultivated diploid Scutellaria baicalensis in Gansu province.Methods:Using ultra performance liquid chromatography(UPLC) to set up the fingerprint,and to simultaneously determine nine components in Scutellaria baicalensis.Results:The similarity index of new strain D20 tetraploid and diploid were greater than 0.99,The contents of baicalin,wogonoside,baicalein,wogonin,scutellarin,luteolin,apigenin,apigenin-7-O-β-D-glucuronide and oroxylin-A of diploid were (14.72 ± 0.46) %,(5.84 ± 0.31) %,(1.55 ± 0.49) %,(0.72 ± 0.21) %,(0.81 ± 0.04) %,(1.47 ± 0.17) %,(0.55 ± 0.01) %,(0.30 ± 0.04) %,(0.12 ± 0.01) %,respectively,while in tetraploid the results were (16.54 ± 1.45)%,(6.62 ±0.39)%,(0.28 ±0.01)%,(0.20 ±0.02)%,(0.87 ±0.06)%,(1.50±0.19)%,(0.19 ±0.04)%,(0.31 ± 0.01)%,(0.10 ± 0.04)%,respectively.Conclusion:The results show that different ploidy have almost the same essential chemical constituent but the different relative contents.Except the baicalein,wogonin,oroxylin-A and apigenin of tetraploid were lower than those of diploid,the others five components of the tetraploid are higher than the diploid.
