摘要:目的观察热补针法对类风湿关节炎(RA)大鼠关节滑膜细胞JAK-STAT信号通路SOCS1、SOCS3表达的影响,探讨热补针法治疗RA的作用机制。方法将48只雄性SD大鼠随机分为正常组、模型组、平补平泻组、热补针法组,每组12只。除正常组外,其余大鼠制备胶原诱导性关节炎(CIA)大鼠模型。成模后平补平泻组和热补针法组取双侧“足三里”,前者施平补平泻法,后者施热补针法,持续1 min,每日1次,共治疗7次,每次留针30 min。测量各组大鼠足趾容积和足底痛反应时间;免疫荧光染色和Western blot检测关节滑膜组织SOCS1、SOCS3的表达。结果与正常组比较,模型组大鼠关节肿胀,足趾容积明显增加,足底痛反应时间明显缩短,关节滑膜组织SOCS1、SOCS3表达下降(P<0.05);与模型组比较,平补平泻组和热补针法组大鼠足趾容积明显减少,足底痛反应时间明显延长,关节滑膜组织SOCS1、SOCS3表达明显升高(P<0.05),且热补针法组均优于平补平泻组(P<0.05)。结论热补针法通过提高模型大鼠滑膜细胞JAK-STAT信号通路细胞因子SOCS1、SOCS3表达,达到抗炎效应及调整滑膜细胞分泌功能的作用。
Objective To observe the effects of heat-reinforcing needling on the expressions of SOCS1 and SOCS3 in JAK-STAT signal pathway of synovial cells of rheumatoid arthritis(RA)rats;To further explore the mechanism of heat-reinforcing needling on RA.Methods A total of 48 male SD rats were randomly selected as normal group,model group,uniform reinforcing-reducing method group,and heat-reinforcing needling group,with 12 cases in each group.Except for the normal group,the rats in other groupswere used to prepare collagen induced arthritis(CIA)rat model.After the model was established,Zusanli acupoint was taken from uniform reinforcing-reducing method group and heat-reinforcing needling group;the former received uniform reinforcing-reducing method,while the latter received heat-reinforcing needling,for 1 min,once a day,for 7 times,the needle being left for 30 min each time.The toe volume and plantar pain response time of each group of rats were measured;the expressions of SOCS1 and SOCS3 in synovial tissue of joints were detected by immunofluorescence staining and Western blot.Results Compared with the normal group,the swelling volume of the joints in the model group significantly increased,the reaction time of plantar pain was significantly shortened,and the expressions of SOCS1 and SOCS3 in synovium of joints decreased(P<0.05).Compared with the model group,the swelling volume of the joints of the rats in uniform reinforcing-reducing method group and heat-reinforcing needling group was significantly reduced,the reaction time of plantar pain was significantly prolonged,and the expressions of SOCS1 and SOCS3 in synovium of joints increased(P<0.05).The swelling volume,pain response time,SOCS1 and SOCS3 expressions in the heat-reinforcing needling group were all better than those in the uniform reinforcing-reducing method group(P<0.05).Conclusion Heat-reinforcing needling can increase the expressions of cytokines SOCS1 and SOCS3 in the JAK-STAT signaling pathway of model rat synovial cells,thereby achieving anti-inflammatory effects and regulating the secretory function of synovial cells.
