摘要:目的筛选甲状腺乳头状癌(papillary thyroid carcinoma,PTC)显著差异表达的microRNA(miRNA),分析其与PTC发病机制的相关性。方法选取确诊的PTC癌组织及癌旁正常组织样本各6份,提取总RNA,采用转录组测序技术,筛选显著差异表达的miRNA,预测其靶基因并进行基因功能(Gene Ontology,GO)注释及信号通路数据库(Kyoto Encyclopedia of Genes and Genomes,KEGG)分析;qRT-PCR法验证显著差异的miRNA在正常人甲状腺细胞(Nthyori3-1)及人甲状腺乳头状癌细胞(TPC-1及k1)中的表达水平。结果共测得2036个miRNA,已知1591个,新发现445个;共发现1166个miRNA差异表达,表达上调446个,表达下调720个,其中miR-124、mi R-7显著下调,miR-494、miR-106a-3p及novel-miR-26显著上调。经GO及KEGG分析,这5个显著差异miRNA的靶基因参与细胞周期、迁移及凋亡,与AMPK、Axon guidance、氨基酸代谢等信号通路的激活关系密切。novel-miR-26在TPC-1及k1细胞中表达量较在Nthy-ori3-1细胞中相对高表达(P均<0.01);miR-124在TPC-1及K1细胞中的表达量较在Nthyori3-1中相对低表达(P均<0.01)。结论显著差异表达的miR-124、miR-494、miR-106a-3p、miR-7及novel-miR-26可作为PTC潜在的分子标志物;Axon guidance、AMPK及蛋白代谢等信号通路在PTC的调控机制中起重要作用。
Objective To screen the miRNAs with significantly differential expression of thyroid papillary carcinoma(PTC)and analyze its correlation to the pathogenic mechanism of PTC.Methods Six confirmed PTC tissue samples and six adjacentnormal tissue samples were collected,from which the total RNAs were extracted and analyzed by transcriptome sequencing to identify the significantly differentially expressed mi RNAs,while the target genes were predicted and subjected to Gene Ontology(GO)annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis.The expression levels of significantly differential miRNAs in normal human thyroid(Nthy-ori3-1)cells and PTC(TPC-1 and k1)cells were determined by qRT-PCR.Results A total of 2036 miRNAs were obtained,of which 1591 were known while 445 were newly discovered.A total of 1166 miRNAs were differentially expressed,with 446 expressions up-regulated and 720 expressions down-regulated.In-depth analysis revealed that the expressions of miR-124 and miR-7 were down-regulated significantly,while those of miR-494,miR-106 a-3p and novel-miR-26 were significantly upregulated.GO and KEGG analyses showed that the five miRNAs involved in cell cycle,cell migration and apoptosis,and were closely related to the activation of signaling pathways such as AMPK,Axon guidance and amino acid metabolism.The expression levels of novel-miR-26 in TPC-1 and k1 cells were higher than that in Nthy-ori3-1 cells(each P<0.01).However,the expression levels of miR-124 in TPC-1 and k1 cells were lower than that in Nthy-ori3-1 cells(each P<0.01).Conclusion Significantly differentially expressed miR-124,miR-494,miR-106a-3p,miR-7 and novel-miR-26 may be used as potential molecular markers of PTC.The signaling pathway such as Axon guidance,AMPK and protein metabolism plays an important role in the regulation mechanism of PTC.
