摘要:目的 通过生物信息学方法研究溶血卵磷脂胆碱酰基转移酶1(LPCAT1)基因在胶质瘤与瘤旁组织中的差异表达,分析LPCAT1基因不同表达水平与胶质瘤患者生存期及预后的关系,并对相关基因进行信号通路富集分析,从而为研究胶质瘤的发生、发展及预后标志物提供依据。方法 通过生物信息学在线软件UALCAN和GEPIA分析、研究LPCAT1基因在胶质瘤与瘤旁组织中差异表达的倍数,并采用UALCAN软件在线分析与LPCAT1差异表达具有相关性的基因,采用DAVID 6.8软件对与LPCAT1具有相关性的基因进行京都基因与基因组百科全书(KEGG)信号通路富集分析,采用Human Protein Atlas在线分析软件进行胶质瘤LPCAT1基因不同表达水平患者的生存期分析,采用STRING数据库和Cytoscape 3.8软件对KEGG聚类分析中发现的与肿瘤密切相关的基因进行关键核心基因筛选。结果 LPCAT1是胶质瘤中高表达的癌基因。与LPCAT1基因具有相关性的上调基因共2486个,具有相关性的下调基因共299个,富集在73个信号通路中,富集倍数大于2的信号通路有33个,其中与肿瘤关系密切的有11个,分别为肾细胞癌、急性白血病、子宫内膜癌、ErbB信号通路、Notch信号通路、Hedgehog信号通路、缺氧诱导因子-1(HIF-1)信号通路、慢性白血病、结肠癌、m TOR信号通路、甲状腺癌。生存期分析结果显示,与胶质瘤LPCAT1基因低表达组比较,高表达组患者的生存期明显缩短(P <0.05),预后相对较差。GRB2、RPS6、STAT3、NOTCH1、GSK3B、MTOR、PTK2、STAT5B、CCND1、GAB1、VEGFA、NFKB1、TSC2、JUN、RELA、ETS1是参与LPCAT1调控胶质瘤生物表型的关键核心基因。结论 LPCAT1参与了胶质瘤的发生、发展,有望成为判断胶质瘤患者预后的肿瘤标志物;通过生物信息学分析发现的与LPCAT1具有高度相关性的癌基因和信号通路可为探索LPCAT1基因的功能和分子机制提供线索和依据。
Objective To study the differential expression of lysophosphatidylcholine acyltransferase 1( LPCAT1) gene between glioma and adjacent tissues by bioinformatics,to analyze the relationship between the different expression levels of LPCAT1 gene and the survival and prognosis of glioma patients and conduct signal pathway enrichment analysis of related genes,thereby providing a reference for the exploration of the occurrence and development of gliomas and the prognosis markers. Methods Through the bioinformatics online software UALCAN and GEPIA,we analyzed and studied the multiple of the differential expression of LPCAT1 gene in glioma and adjacent tissues,and used the software UALCAN to analyze the genes related to the differential expression of LPCAT1 online,and used DAVID 6. 8 software to conduct the signal pathway enrichment analysis of Kyoto encyclopedia of genes and Genomes( KEGG) for the genes related to LPCAT1,used Human Protein Atlas online analysis software to analyze the survival of patients with different expression levels of glioma LPCAT1 gene,and used STRING database and Cytoscape 3.8 software to screen out the key core genes found in KEGG cluster analysis that are closely related to tumors. Results LPCAT1 is a highly expressed oncogene in glioma. There were 2486 up-regulated genes related to LPCAT1 gene,299 down-regulated genes related to it,which were enriched in 73 signal pathways,and 33 signal pathways with fold enrichment greater than 2,among which 11 were closely related to tumors,including renal cell carcinoma,acute leukaemia,endometrial carcinoma,ErbB signaling pathway,Notch signaling pathway,Hedgehog signaling pathway,hypoxia inducible factor-1( HIF-1) signaling pathway,chronic leukemia,colon cancer,m TOR signaling pathway and thyroid cancer. The survival period analysis showed that the survival period of patients in the high expression group of glioma LPCAT1 gene significantly shortened as compared with the low expression group thereof( P<0.05),and the prognosis was relatively poor. GRB2,RPS6,STAT3,NOTCH1,GSK3B,MTOR,PTK2,STAT5B,CCND1,GAB1,VEGFA,NFKB1,TSC2,JUN,RELA and ETS1 were key core genes involved in the regulation of glioma biological phenotype by LPCAT1. Conclusion LPCAT1 is involved in the occurrence and development of glioma and is expected to become a tumor marker to judge the prognosis of glioma patients;The oncogenes and signal pathways with high correlation with LPCAT1 discovered by bioinformatics analysis can provide clues and references for exploring the function and molecular mechanism of LPCAT1 gene.
