摘要:目的:研究消定膏(无名异、儿茶、酒大黄、紫荆皮)对兔骨折愈合中血清骨钙素、骨痂ATPase a3不同时相表达的影响,探讨其对骨折愈合的作用机制.方法:将128只新西兰大耳白兔随机分为4组:空白组、模型组、青鹏膏组(阳性对照组)和消定膏组,除空白组外,其余3组均采用骨缺损法造成胫骨3 mm骨缺损模型,造模成功后,阳性对照组外敷青鹏膏、消定膏组外敷消定膏,分别于用药后第3、7、14、28天4个不同时相点各组随机抽取8只白兔,X光拍片后比较骨痂生长及骨折愈合情况,心脏取血,酶联免疫吸附(ELISA)法检测血清骨钙素水平;取白兔骨痂组织,光镜下观察病理形态学变化,免疫组化法检测骨痂中ATPase a3蛋白表达情况.结果:与模型组、青鹏膏组比较,消定膏组白兔骨痂生长及评分明显优于其他组,消定膏组血清骨钙素和骨痂ATPase a3蛋白免疫组化平均光密度值在用药后逐渐增高,第7~14天左右达到峰值,随后下降(P<0.05).结论:消定膏对兔胫骨骨折愈合的促进作用可能与其调控血清骨钙素和骨痂ATPase a3蛋白有关.
Objective:To study the effect of the Xiaoding Ointment on the expression of osteocalcin and ATPase a3 during different stages of fracture healing in a rabbit model,and to explore the mechanism of the treatment effect.Methods:One hundred and twenty-eight New Zealand white rabbits were randomly divided into blank group、model group、Qingpeng Ointment group(as the positive control group)and Xiaoding Ointment group.Except the blank group,all the other groups were made tibia 3 mm bone defect model using bone defecting method.After the success of the modeling,positive control group was treated with Qingpeng Ointment、Xiaoding Ointment group was treated with Xiaoding Ointment respectively.After 3,7,14,and 28 d,eight rabbits were randomly selected from each group.X-ray radiography was used to compare the growth of callus and fracture healing,enzyme-linked immuno sorbent assay(ELISA)were used to detect serum osteocalcin in blood sampling from the heart,the callus tissue was collected and the morpho pathology changes of the callus were observed under an optical microscope,the expression of ATPase a3 protein in callus tissue was detected by immunohistochemistry.Results:Compared with the model group and the Qingpeng Ointment group,the callus growth and grading results of the Xiaoding Ointment group was significantly better than that of the other groups.The expressions of osteocalcin and ATPase a3 in samples from Xiaoding Ointment group were gradually increased after surgery,which reached peak around 7 to 14 d(P<0.05).Conclusion:The regulation of osteocalcin and ATPase a3 is one of the potential mechanism underlying the promotion of Xiaoding Ointment on fracture healing.
