摘要:目的:探讨当归挥发油对人肺腺癌SPC-A-1细胞增殖及凋亡的作用。方法:MTT比色法检测当归挥发油对SPC-A-1细胞的细胞毒作用;流式细胞术检测SPC-A-1细胞凋亡及周期;吖啶橙/溴化乙啶(AO/BE)双荧光染色法观察SPC-A-1细胞凋亡。结果:在6.25-400μg/ml的浓度范围内,当归挥发油对SPC-A-1细胞的增殖具有浓度依赖性抑制作用,其对SPC-A-1细胞的IC50为112.38μg/ml。流式细胞术检测结果表明,当归挥发油可诱导SPC-A-1细胞凋亡、坏死,且细胞凋亡、坏死率随药物浓度增高而升高。AO/BE双荧光染色法结果显示,用药组随着药物浓度的升高,细胞结构固缩加重,凋亡、坏死细胞增多。结论:当归挥发油可诱导SPC-A-1细胞凋亡。
Objective: To investigate the cytotoxic effect and apoptosis of volatile oil of the angelica on human lung adenocarcinoma cell SPC-A-I. Methods: The cytotoxic effect of human lung adenocarcinoma cell SPC-A-1 was investigated by MTT colorimetric assay. The cell apoptosis and cell cycle were analyzed by flow cytometry. The cell apoptosis was observed by AO/BE fluorescent staining method. Results: The SPC- A-1 cell was inhibited by volatile oil of the angelica in a concentration-dependent manner, the IC50 was 112.38 μg/ml. The result of flow cy-tometry indicate that, volatile oil of the angelica could induced apoptosis and necrosis of SPC-A-1 cell, and the rate of apoptosis and necrosis all rose with the raise of drug concentration. The results of AO/BE fluorescent staining method displayed that, the rates of apoptosis and nec- rosis all rose in drug groups. The result of flow eytometry indicated that, after using volatile oil of the angelica, the SPC-A-1 cell cycle was blocked in G2. Conclusion : Volatile oil of the angelica had depressant effect on the generation of SPC-A-1 cell, the mechanism is probably that it can induce apoptosis of SPC-A-1 cell.
