摘要:目的观察敦煌医学古方大补脾汤对放射性肺损伤大鼠PI3K/Akt信号通路的影响,探讨其作用机制。方法40只SPF级SD大鼠随机分为空白对照组、模型组、单次给药组、两次给药组,每组10只。各给药组每日予大补脾汤灌胃,空白对照组和模型组予等量生理盐水灌胃。2周后,模型组、单次给药组、两次给药组大鼠右肺予8 Gy X射线单次照射,空白对照组不予照射。照射后,两次给药组继续每日予大补脾汤灌胃,其余各组灌胃等量生理盐水。2周后处死所有大鼠,ELISA检测大鼠血清PI3K、Akt、p-PI3K、p-Akt含量,Western blot及免疫组化检测大鼠右肺组织PI3K、Akt、p-PI3K、p-Akt蛋白表达,qPCR检测大鼠右肺组织PI3K、Akt mRNA表达。结果与空白对照组比较,模型组大鼠血清PI3K、Akt、p-PI3K、p-Akt含量明显增加(P<0.01),右肺组织PI3K、Akt、p-PI3K、p-Akt蛋白及PI3K、Akt mRNA表达明显升高(P<0.01);与模型组比较,单次给药组和两次给药组大鼠血清PI3K、Akt、p-PI3K、p-Akt含量明显减少(P<0.05,P<0.01),右肺组织PI3K、Akt、p-PI3K、p-Akt蛋白及PI3K、Akt mRNA表达显著降低(P<0.05,P<0.01)。结论大补脾汤对放射性肺损伤大鼠具有保护作用,其机制与抑制PI3K、Akt磷酸化,调控PI3K/Akt信号通路相关。
Objective To observe the effects of Dabupi Decoction of ancient prescription of Dunhuang medicine on PI3K/Akt signaling pathway in rats with radiation-induced lung injury;To explore the possible mechanism.Methods Totally 40 SPF SD rats were randomly divided into four groups:normal control group,model group,single-dosage group and twice-dosage group.The single-dosage group and twice-dosage group were given Dabupi Decoction for gavage daily,and the normal control group and the model group were given the equal normal saline for gavage.After two weeks,the right lungs of the rats of model group,single-dosage group and twice-dosage group were radiated once by 8 Gy X-ray,while the normal control group was not radiated.After radiation,Dabupi Decoction was continued for gavage daily in the twice-dosage group.2 weeks after radiation,all groups of rats were executed.The contents of PI3K,Akt,p-PI3K and p-Akt in rat serum were detected by ELISA.The protein expressions of PI3K,Akt,p-PI3K and p-Akt in the right lung tissue of rats were detected with Western blot and immunohistochemistry staining,and mRNA expressions of PI3K and Akt in the right lung tissue of rats was detected with qPCR.Results Compared with the normal control group,the contents of PI3K,Akt,p-PI3K and p-Akt in rat serum of the model group significantly increased(P<0.01),and the protein expressions of PI3K,Akt,p-PI3K and p-Akt and mRNA expressions of PI3K and Akt in the right lung tissue were significantly up-regulated(P<0.01).Compared with the model group,the contents of PI3K,Akt,p-PI3K and p-Akt in rat serum of the single-dosage group and twice-dosage group significantly decreased(P<0.05,P<0.01),the protein expressions of PI3K,Akt,p-PI3K and p-Akt in the right lung tissue significantly decreased and the mRNA expressions of PI3K and Akt also decreased(P<0.05,P<0.01).Conclusion Dabupi Decoction has protective effect on rats with radiation-induced lung injury,and its mechanism is related to regulating PI3K/Akt signaling pathway.
