摘要:目的:观察红芪多糖对实验性脾虚大鼠胃黏膜组织中COX-2表达的影响。方法:100%大黄水煎液连续42天灌胃制备脾虚证大鼠模型,造模后将大鼠随机分为6组,其中空白对照组、模型对照组正常饲养,药物组(高、中剂量组)用红芪多糖灌胃30天,对照组(高、中剂量组)以黄芪多糖灌胃30天进行干预;用免疫组化法检测大鼠胃黏膜组织中环氧合酶-2(cycloxygenase-2,COX-2)的表达率。结果:模型对照组COX-2阳性表达率与空白组比较显著升高(P<0.01),红芪多糖高剂量组,黄芪多糖高剂量组COX-2阳性表达率与模型组比较显著降低(P<0.01),红芪多糖中剂量组COX-2阳性表达率与模型组比较明显降低(P<0.05)。结论:红芪多糖能够影响脾虚大鼠胃黏膜中病理因子COX-2的表达,能提高黏膜修复能力,从而防止脾虚证胃黏膜发生进一步病变。
Objective:To observe the influence of radix hedysari polysaccharide to expression of COX-2 in gastric mucosa of rat with experimental PiXu.Method:Rats are given intragastric infusion of 100% water decoction of DaHuang continually for 42 days in order to make the models with PiXu.Then the rats are randomly divided into six groups.Rats in model group receive normal feeding,the ones in medication group(high-dose group and middle dose group) are given intragastric infusion of radix hedysari polysaccharide for 30 days and the ones in control group given intragastric infusion of astragalus polysaccharide intervene for 30 days.The expression rates of COX-2 in gastric mucosa of rat are detected with immunohistochemical method.Result:The positive rates of COX-2 of model control group are significantly higher than blank group.To compare with model group,the positive rates of high-dose radix hedysari polysaccharide group,high-dose astragalus polysaccharide group are significantly decreased(P0.01),while of middle dose radix hedysari polysaccharide group are obviously decreased(P0.05).Conclusion:Radix hedysari polysaccharide can influence the expression of pathological factor COX-2 in gastric mucosa of rats and improve the repair of mucosa,so that it can prevent further pathological changes of gastric mucosa in PiXuZheng.
