摘要:目的:基于数据挖掘分析中医药治疗新型冠状病毒感染(简称新冠感染)初期的用药规律,同时采用网络药理学分析高频药物治疗新冠感染初期的潜在作用机制。方法:检索中国知网、万方、维普中文期刊和中国生物医学文献数据库中2019年12月12日―2022年12月10日中医或中西医结合治疗新冠感染初期的文献,对文献中所涉及的方剂进行筛选、整理,采用R语言对药物组成进行频数分析、关联规则分析及聚类分析;从中药系统药理学数据库与分析平台(TCMSP)数据库和GeneCadrs等数据库中分别获得药物和新冠感染相关靶点,利用Venny取交集后通过Cytoscape 3.9.1制作药物-成分-靶点网络;将共同靶点上传STRING数据库进行蛋白质-蛋白质相互作用(Protein-protein Interaction,PPI)网络分析;运用Metascape数据库对共同靶点进行京都基因与基因组百科全书(KEGG)通路和基因本体论(GO)功能富集分析。结果:共收集到中医药治疗新冠感染初期的处方42首,高频药物有苦杏仁、广藿香、茯苓、麻黄、半夏、黄芩等17味中药;关联规则分析关联系数高的药物组合分别为半夏-麻黄-桂枝、半夏-苦杏仁-麻黄-桂枝;聚类分析将药物分为4类;对前五味高频药物苦杏仁、广藿香、茯苓、麻黄、半夏进行网络药理学分析,获取高频药物与新冠感染交集靶点62个,核心靶点6个:丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、肿瘤蛋白p53(Tumor Protein p53,TP53)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)等,涉及通路165条,主要包括T细胞受体信号通路、白细胞介素-17信号通路等。结论:中医药治疗新冠感染初期用药多以清热解毒、宣肺平喘、燥湿化痰、补气健脾为主,其高频药物主要通过参与炎症反应、免疫调节等过程治疗新冠感染初期,为中医药治疗新冠感染初期提供理论依据。
Objective:To analyze the medication regularity of traditional Chinese medicine in the treatment of novel coronavirus infection at the initial stage based on data mining and to analyze the potential action mechanism of high-frequency drugs in the treatment of novel coronavirus infection at the initial stage by network pharmacology.Methods:Literature on the treatment of the initial stage of the novel coronavirus infection in traditional Chinese medicine or integrated TCM and Western medicine was retrieved from CNKI,Wangfang,VIP Chinese journals and CBM Database from December 12,2019 to December 10,2022,the prescriptions involved in the literature were screened and sorted out,and the frequency analysis,association rule analysis and cluster analysis of drug composition were carried out by R language.Targets related to drug and novel coronavirus infection were obtained from TCMSP database and GeneGards database,respectively.After intersection by Venny,Cytoscape 3.9.1 was used to make drug-ingredient-target network,and the common target was uploaded to the STRING database for a PPI analysis.The Metascape database was used to analyze the KEGG pathway and GO function enrichment of common targets.Results:A total of 42 prescriptions of traditional Chinese medicine were collected for the treatment of the initial stage of the novel coronavirus infection,and 17 kinds of traditional Chinese medicine were frequently used,including Kuxingren(Bbitter almond),Guanghuoxiang(Patchouli),Fuling(Poria cocos),Mahuang(Ephedra),Banxia(Pinellia pinellia)and Huangqin(Scutellaria scutellaria).The drug combinations with high association coefficients were Banxia-Mahuang-Guizhi(Cassia twig),Banxia-Kuxingren-Mahuang-Guizhi,respectively.Cluster analysis divided drugs into four groups.The network pharmacological analysis of the first five high frequency drugs,Kuxingren,Guanghuoxiang,Fuling,Mahuang,Banxia were carried out,a total of 62 intersection targets of high-frequency drugs and novel coronavirus infection were obtained,and there were 6 core targets,such as AKT1,TP53,and TNF,involving 165 pathways,including T cell receptor signaling pathway and interleukin-17 signaling pathway.Conclusion:At the initial stage of novel coronavirus infection,TCM mainly applies the methods of clearing heat and detoxifying,promoting lung and relieving asthma,drying dampness and eliminating phlegm,and invigorating Qi(气)and spleen;high-frequency drugs are mainly involved in the process of inflammation and immune regulation to treat the initial stage of novel coronavirus infection,providing a theoretical basis for TCM treatment of the initial stage of novel coronavirus infection.
