摘要:目的探讨敦煌固本方对腹泻型肠易激综合征(Diarrhea-predominant irritable bowel syndrome,D-IBS)大鼠血清白细胞介素-6(IL-6)和5-羟色胺(5-HT)水平及结肠组织水通道蛋白3(AQP3)、核因子-κB(NF-κB)表达的影响。方法采用束缚联合番泻叶灌胃的方法建立D-IBS大鼠模型,并进行结肠转运功能检测。将50只雌性SD大鼠随机分为正常组、模型组、阳性药物组(补脾益肠丸组,0.8 g·kg-1·d-1)、敦煌固本方低剂量组(0.6 g·kg-1·d-1)、敦煌固本方高剂量组(1.2 g·kg-1·d-1)。每日1次,连续灌胃给药40 d后,采集大鼠血清及结肠组织标本。采用ELISA法检测大鼠血清IL-6、5-HT水平;qRT-PCR法检测大鼠结肠组织AQP3 mRNA表达水平;Western Blot法检测大鼠结肠组织AQP3蛋白表达;免疫组化法检测大鼠结肠组织NF-κB蛋白表达。结果与正常组比较,D-IBS模型组大鼠从第7天开始,粪点数显著增加(P <0.01),玻璃小球推出时间显著降低(P <0.01);血清IL-6、5-HT及NF-κB蛋白表达水平显著升高(P <0.01),结肠组织AQP3 mRNA及蛋白表达水平显著降低(P <0.01)。与模型组比较,敦煌固本方低剂量组的大鼠血清IL-6水平显著降低(P <0.01);敦煌固本方高剂量组的大鼠血清IL-6、5-HT以及NF-κB蛋白表达水平显著降低(P <0.01),结肠组织AQP3 mRNA及蛋白表达水平显著升高(P <0.01)。结论敦煌固本方可能通过降低D-IBS大鼠血清IL-6、5-HT以及NF-κB表达水平,提高结肠组织AQP3表达,从而缓解D-IBS大鼠腹泻等症状。
Objective To investigate the effect of Dunhuang Guben recipe on serum levels of interleukin-6(IL-6),5-hydroxytryptamine(5-HT),and aquaporin 3(AQP3),nuclear factor-kappa B(NF-κB)expression in colon tissue of diarrhea-predominant irritable bowel syndrome(D-IBS)rats.Methods D-IBS rat model was established by the restraint and senna treatment.Fifty female SD rats were randomly divided into normal group,model group,positive control group(Bupi Yichang pills treatment group,0.8 g·kg-1·d-1),low dose Dunhuang Guben recipe group(0.6 g·kg-1·d-1)and high dose Dunhuang Guben recipe group(1.2 g·kg-1·d-1).After drug treatment of continuous administration for 40 days,rats’serum and colon tissue samples were collected.Serum levels of IL-6 and 5-HT were measured by ELISA.The expression of AQP3 mRNA in colonic tissue was detected by qRT-PCR.The expression of AQP3 protein in colonic tissue was detected by Western Blot,and the expression of NF-κB protein in colon tissue was detected by immunohistochemistry.Results Compared with the normal group,the number of fecal points were increased significantly(P<0.01),and the ejection time of glass pellets were decreased significantly(P<0.01)from 7 th day in D-IBS model rats.Serum levels of IL-6 and 5-HT in D-IBS model rats were significantly increased(P<0.01);the expression of AQP3 mRNA and AQP3 protein in colon tissue of D-IBS model rats were significantly decreased(P<0.05).Compared with the model group,serum levels of IL-6 in the low dose Dunhuang Guben recipe group were significantly decreased(P<0.01);serum levels of IL-6,5-HT and colon tissue protein level of NF-κB in the high dose Dunhuang Guben recipe group were significantly decreased(P<0.01).The expression of AQP3 mRNA and protein in colon tissue of the high dose Dunhuang Guben recipe group were significantly increased(P<0.01).Conclusion Dunhuang Guben recipe could alleviate diarrhea and other symptoms in D-IBS rats by decreasing the expression of IL-6,5-HT and NF-κB in D-IBS rats,and increasing the expression of AQP3 in colon tissue.
