摘要:目的探讨三黄膏对耐甲氧西林金黄色葡萄球菌(MRSA)感染大鼠皮肤软组织的干预作用。方法将96只Wistar大鼠随机分为空白组、模型组、对照组和高、中、低剂量实验组,每组16只。除空白组外,其余大鼠行皮下软组织注射MRSA菌悬液造模。造模成功后,高、中、低剂量实验组分别给予三黄膏1.00、0.50和0.25 g外敷于患处,空白组和模型组均涂抹等量凡士林(1.00 g),对照组给予莫匹罗星0.50 g。6组大鼠每12 h换药1次,连续给药7 d。治疗7 d后处死大鼠,计算创面愈合率,用实时荧光定量聚合酶链反应检测感染组织中白细胞介素-1受体相关激酶1(IRAK-1)和IκB激酶β(IKKβ)mRNA的表达水平,用酶联免疫吸附法检测白细胞介素-6(IL-6)和IL-8的含量。结果治疗7 d后,中、高剂量实验组和对照组、模型组的创面愈合率分别为(88.63±4.54)%、(93.98±3.45)%、(83.02±5.87)%和(60.30±2.30)%,对照组和中、高剂量实验组的创面愈合率与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。高剂量实验组、对照组、模型组和空白组的IRAK-1 mRNA分别为1.14±0.04、1.18±0.05、1.42±0.05和1.01±0.05,IKKβmRNA分别为1.28±0.04、1.30±0.05、1.62±0.04和1.00±0.04,血清IL-6含量分别为(17.83±0.26)、(17.48±0.14)、(19.32±0.42)和(14.51±0.24)pg·mL^(-1),血清IL-8含量分别为(29.04±0.94)、(29.12±0.90)、(33.13±1.01)和(26.37±1.02)pg·mL^(-1)。高剂量实验组和对照组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论三黄膏可能通过下调IRAK-1和IKKβ表达,抑制下游炎性分子IL-6和IL-8释放,进而发挥抗炎作用,促进感染创面愈合。
Objective To investigate the effect of Sanhuang ointment on skin and soft tissue of rats infected with methicillin-resistant Staphylococcus aureus(MRSA).Methods A total of 96 Wistar rats were randomly divided into blank,model,control and experimental-L,-M,-H groups,with 16 rats per group.Except for the blank group,other rats were injected with MRSA bacteria suspension subcutaneously to make model.After success,experimental-L,-M,-H groups were given Sanhuang ointment 1.00,0.50 and 0.25 g to the affected area.Vaseline 1.00 g of the same amount was applied to the blank and model groups,and the control group was given mupirocin 0.50 g.Six groups of rat s c hanged their dressing every 12 hours for 7 consecutive days.After 7 days of treatment,the rats were killed,the wound healing rate was calculated,and the mRNA expression levels of interleukin-1 receptor-associated kinase 1(IRAK-1)andκB kinaseβ(IKKβ)in infected tissues were detected by real-time fluorescence quantitative polymerase chain reaction,the contents of interleukin-6(IL-6)and IL-8 were detected by enzyme-linked immunosorbent assay.Results After 7 days of treatment,the wound healing rates of the wound healing rates of experimental-L,-M,-H groups,control group and model group were(88.63±4.54)%,(93.98±3.45)%,(83.02±5.87)%and(60.30±2.30)%,respectively.The wound healing rates of the control group,experimental-M,-H groups were significantly different from that of model group(P<0.05,P<0.01).The expression levels of IRAK-1 mRNA in the experimental-H,control,model and blank groups were 1.14±0.04,1.18±0.05,1.42±0.05 and 1.01±0.05;the expression levels of IKKβmRNA were 1.28±0.04,1.30±0.05,1.62±0.04 and 1.00±0.04;the serum IL-6 contents were(17.83±0.26),(17.48±0.14),(19.32±0.42)and(14.51±0.24)pg·mL^(-1);and the serum IL-8 contents were(29.04±0.94),(29.12±0.90),(33.13±1.01)and(26.37±1.02)pg·mL^(-1).Comparing with the model group,differences of the above indexes in the experimental-H and control groups were statistically significant(P<0.05,P<0.01).Conclusion Sanhuang ointment may inhibit the release of downstream inflammatory molecules IL-6 and IL-8 by down-regulating the expressions of IRAK-1 and IKKβ,thereby exerting anti-inflammatory effects and promoting wound healing.
