摘要:目的探讨不同补肾法诱导的骨髓间充质干细胞(BMSCs)对脑梗死大鼠神经功能恢复的影响。方法用线栓法制备大鼠大脑中动脉梗死(MCAO)模型,随机分为假手术组、模型组、单纯BMSCs移植组及左归丸、右归丸、地黄饮子、维甲酸诱导BMSCs移植组。移植后3、7、14 d进行大鼠神经功能综合测评,于移植后7、14 d处死大鼠,采用TTC染色法测定脑梗死面积,并采用免疫组化方法检测脑组织神经突触素(syn)的表达。结果与模型组比较,各移植组7、14 d的神经功能评分均显著增高(P<0.05),脑梗死面积均显著缩小(P<0.05),syn阳性细胞数均明显增加(P<0.05)。结论不同补肾法诱导BMSCs能够促进脑梗死大鼠神经功能的恢复,且补阴和阴阳双补方药诱导BMSCs组的作用优于补阳类方药。
Objective To explore neural functional recovery of cerebral infarction rats promoted by bone marrow mesenchymal stem cells (BMSCs) induced by kidney-nourishing methods. Methods Middle cerebral artery occlusion (MCAO) model was made and rats were randomly divided into psudo-operation group, model group, BMSCs transplantation group and Zuoguiwan-, Youguiwan-, Dihuangyinzi-, retinoic acid-induced BMSCs transplantation group. On the 3rd, 7th, 14th day after transplantation, neural function of the rats was comprehensively tested. On the 7th, 14th day after transplantation, the rats were executed. TTC dying method was used to determine the cerebral infarction area, and imrnunohistochemical method was used to test brain tissue synaptophysin expression. Results Compared with the indexes of model group, on 7th, 14th day, neurofunction score of each transplantation group was significantly increased (P 〈0.05), cerebral infarction area of each transplantation group was significantly reduced (P 〈0.05), and synaptophysin positive cells of each transplantation group were significantly increased (P 〈0.05). Conclusion BMSCs induced by different kidney-nourishing method can promote neural function recovery of cerebral infarction rats. The effects of yin-nourishing and yinyang-nourishing prescriptions are better than yang-nourishing prescription.
