摘要:目的观察当归挥发油对自发性高血压大鼠的血压、肾素及血管紧张素Ⅱ(AngⅡ)表达的影响,探讨其可能的降压机制。方法采用随机数字表将自发性高血压大鼠分为模型组、当归挥发油不同剂量组和缬沙坦组,将同龄Wistar大鼠作为正常对照组,每组10只。给予不同给药组大鼠相应浓度的药物,给予模型组和正常组大鼠等量生理盐水,ig给药4周,测定给药前、后大鼠尾动脉的收缩压;给药4周后,麻醉大鼠,取出心肌组织,采用HE染色观察心肌组织细胞的形态改变;于大鼠腹主动脉采血,用酶联免疫吸附法检测大鼠血清中肾素、AngⅡ的水平。结果不同给药组大鼠的收缩压均降低,与模型组比较,差异具有统计学意义;HE染色显示:缬沙坦和不同剂量当归挥发油对大鼠的心肌横纹均有不同程度的改善,能抑制心肌细胞肥大,改善心肌细胞的形态和结构;ELISA结果显示:不同给药组大鼠的肾素和AngⅡ水平明显降低。结论当归挥发油具有明显的降压效果,对心肌组织具有保护作用,能够降低血清中肾素和AngⅡ的水平,其降压作用可能与肾素-血管紧张素-醛固酮系统有关。
OBJECTIVE To observe the effect of angelica volatile oil on blood pressure and the expression of renin and angiotensinⅡ(AngⅡ) in spontaneously hypertensive rats(SHR),and to explore its possible mechanism. METHODS The SHR were randomly divided into the model group,different doses of angelica volatile oil group and Valsartan group,and the same age Wistar rats served as the normal control group. Different doses of drugs were given to the rats in different drug delivery groups by gavage for 4 weeks. The same amount of saline were given to the rats in model group and normal group by gavage for 4 weeks. The tail artery systolic pressure of rats were measured before and after administration. After the gavage for 4 weeks. The rats were anesthetized,and the myocardial tissues were taken out and the morphological changes of myocardial cells were observed by HE staining. Abdominal aorta blood were collected,and the levels of renin and angiotensin Ⅱ in the serum were detected by Enzyme-linked immunosorbent method. RESULTS Compared with the model group,the systolic blood pressure of rats in different administration groups were decreased significantly. The myocardial striae of rats in Valsartan and different dosage of angelica volatile oil have got varying degrees improvement according to the HE staining results. Valsartan and different dosage of angelica volatile oil could inhibit cardiac hypertrophy and improve myocardial cell morphology and structure. The levels of renin and angiotensin Ⅱ in the different administration groups were significantly lower.CONCLUSION Angelica volatile oil has a significant antihypertensive effect on myocardial tissue. It has a protective effect and can reduce serum renin and angiotensin Ⅱ levels. Its antihypertensive effect may be related to renin-angiotensin-aldosterone system.
