摘要:目的研究黄芪总苷对氯化镉染毒大鼠肝、肾损伤的保护作用。方法 36只Wistar大鼠随机均分为3组。染毒组和干预组均腹腔注射0.1%氯化镉,每周5次,共5周,染毒的同时干预组给予黄芪总苷(20 mg/kg·bw·d)灌胃治疗,1次/d,连续5周,末次给药24 h后股动脉采血处死大鼠,测定大鼠肝、肾组织的镉含量、SOD活性、LDH活性和MDA含量,观察肝、肾组织的Bcl-2和Bax的蛋白表达,测定大鼠肝组织AST活性、ALT和IL-2含量和TGF-β1的含量,测定大鼠肾组织GSH-Px活性。结果与空白组比较,染毒组肝肾组织镉含量、MDA含量、LDH活性、Bax蛋白表达均明显升高,SOD活性、Bcl-2蛋白表达明显下降(P<0.05)。同时,染毒组肝组织AST活性、ALT活性、TGF-β1含量均明显升高,肝组织IL-2含量和肾组织GSH-Px的活性下降,差异有统计学意义(P<0.05)。与染毒组比较,干预组大鼠肝肾组织、TGF-β1含量、MDA含量、LDH活性、Bax蛋白表达均明显降低,SOD活性、Bcl-2蛋白表达明显升高(P<0.05)。同时,干预组大鼠肝组织AST活性、ALT活性、TGF-β1含量均明显降低,镉含量肾组织GSH-Px的活性明显增强,差异有统计学意义(P<0.05)。结论黄芪总苷对镉所致大鼠的肝肾损伤具有保护作用,其机制可能与黄芪总苷抑制镉染毒大鼠体内氧化应激反应、调控酶活性和细胞周期蛋白有关。
Objective To study the protective effects of astragalosides on liver and renal damage induced by cadmium. Methods Thirty-six wistar rats were randomized divided into three groups. 0. 1% cadmium chloride was intraperitoneally injected into the cadmiumexposed group and the intervention group once a day for five week,at the same time,the intervention group was given astragalosides( 20 mg/kg·bw·d) by gastric lavage. After the last administration,the rats was excuted by femoral artery blood,the levels of cadmium( Cd),superoxide dismutase( SOD),malondialdehyde( MDA),LDH,alanine aminotransferase( AST),aspartate aminotransferase( ALT),glautathione peroxidase( GSH-Px),interleukin-2( IL-2),transforming growth factor-β1( TGF-β1) were checked,the protein expression of Bcl-2 and Bax were also analyzed. Retsults Compared with the blank group,the levels of Cd,LDH,AST,ALT,Bax,TGF-β1 were all increased significantly in the cadmium-exposed group( P〈0.05). But the activities of SOD,IL-2,GSH-Px and the protein expression of Bcl-2 were all decreased obviously in the cadmium-exposed group( P〈0.05). Compared with the cadmium-exposed group,the levels of Cd,MDA,LDH and Bax in liver and renal tissues were all decreased in intervention group( P〈0.05). The levels of AST,ALT and TGF-β1 in liver tissue were reduced in intervention group( P〈0.05). But the activity of SOD in liver and renal tissues was improved in intervention group( P〈0.05),the content of IL-2 in liver tissue was increased in intervention group( P〈0.05),the activity of GSH-Px in renal tissue was also increased in intervention group( P〈0.05). Conclusion Astragalosides has ability to prevent damage of liver and renal in rats that exposed in cadmium,its mechanism could inhibit oxidative stress、regulate enzyme activity and regulate cell cycle protein caused by cadmium.
