摘要:以自发性高血压大鼠(SHR)为研究对象,比较当归-川牛膝水煎剂及不同分离组分对SHR血压的影响。采用D-101型大孔吸附树脂分离当归-川牛膝水煎剂,将试验动物SHR随机分为8组,除模型组外,其余7组分别为缬沙坦组、当归-川牛膝水煎剂组、纯水洗涤组、30%乙醇洗脱组、50%乙醇洗脱组、70%乙醇洗脱组和90%乙醇洗脱组,连续灌胃4周;以相同周龄的Wistar大鼠作为正常对照组,正常对照组和模型组大鼠同步给予相同体积和频次的生理盐水灌胃。采用无创血压检测系统每周测定各组大鼠血压值1次。结果发现,大孔吸附树脂分离当归-川牛膝水煎剂,分离组分中水洗组分占比68.03%,30%乙醇洗脱组分占比2.53%,50%乙醇洗脱组分占比0.60%,70%乙醇洗脱组分占比0.55%,90%乙醇洗脱组分占比0.05%;30%乙醇洗脱组占总醇洗组分的比例最高,达67.73%;各中药治疗组对自发性高血压大鼠血压的治疗效果比较,当归-川牛膝水煎剂的30%乙醇洗脱组分的疗效优于其他分离组分。说明当归-川牛膝水煎剂经大孔吸附树脂分离,所得到的30%乙醇洗脱组分中有机物所占比例最大;当归-川牛膝水煎剂的30%乙醇洗脱组分对血压的干预效果优于其他分离组分。
In this study,spontaneously hypertensive rats(SHR)were taken as the research object. The antihypertensive effects of Angelica sinensis-Achyranthes bidentata decoction and its fractions were analyzed and compared. The effects of Angelica sinensis-Achyranthes bidentata decoction and its fractions on blood pressure were preliminarily explored. In this experiment,D-101 macroporous resin was used to separate Angelica sinensis-Achyranthes bidentata decoction. SHR were randomly divided into eight groups. Except model group,the other seven groups were given valsartan,Angelica sinensis-Achyranthes bidentata decoction,pure water washing component,30% ethanol eluting component,50% ethanol eluting component,70% ethanol eluting component and 90% ethanol eluting component respectively for 4 weeks. Wistar rats of the same age were taken as the control group,normal control group and the model group were given the same volume and frequency simultaneously. The blood pressure of rats in each group was measured once a week before and after treatment by non-invasive blood pressure detection system. 4 weeks after treatment,rats were killed under anesthesia and serum was collected. Macroporous resin was used to separate Angelica sinensis-Achyranthes bidentata decoction. The proportion of water-washing group was 68.03%,that of 30%ethanol-eluting was 2.53%,that of 50% ethanol-eluting was 0.60%,that of 70% ethanol-eluting was 0.55%,and that of 90%ethanol-eluting was 0.05%. The 30% ethanol elution group accounted for the highest proportion of the total ethanol elution components,accounting for 67.73% of the total ethanol elution components. Comparing the therapeutic effects of each Chinese medicine treatment group on blood pressure of SHR,Angelica sinensis-Achyranthes bidentata decoction group and 30% ethanol elution group were better than other separation groups. The result showed when Angelica sinensis-Achyranthes bidentata decoction was separated by macroporous adsorption resin,the proportion of organic matter in the 30% ethanol elution group was the largest. The 30% ethanol elution group of Angelica sinensis-Achyranthes bidentata decoction had better antihypertensive effect than other fractions.
