摘要:目的:建立琼玉润肺膏的质量标准。方法:采用正交设计法以浸泡时间.提取时间、提取次数以及物料比为因素,以中间体中毛蕊花糖苷的含量、中间体中五味子醇甲的含量以及出膏率为指标,优选最佳提取工艺;采用薄层色谱法对制剂中的生地黄、五味子进行定性鉴别;采用高效液相色谱法对本制剂中五味子的主要有效成分五味子醇甲进行含量测定。结果:确定了琼玉润肺膏的最佳提取条件、最佳优选组合为A3B3C1D3,即浸泡4h,加14倍量水,煎煮3次,每次3h.其中生地黄、五味子的薄层色谱鉴别斑点清晰,重现性好,阴性无干扰;确立了本膏方中五味子的主要成分五味子醇甲的HPLC含量测定方法,结果显示五味子醇甲在0.00104~0.0416mg/mL之间线性关系良好,r=0.9998,平均加样回收率为104.56%(6例),RSD=3.48%。结论:本研究建立的方法简便.准确、稳定,可作为琼玉润肺膏的质量控制标准。
Objective: To establish quality standard of Qiongyu Runfei syrup. Methods: Orthogonal design was used to optimize the best extraction process by taking soaking time, extraction time, extraction times and material ratio as the factors, the contents of verbascoside and schizandrol A in intermediate object and extraction ratio as the indexes;TLC was used to identify Sheng Dihuang(Rehmannia Glutinosa Libosch) and Wuweizi(Schisandrae Chinensis Fructus) qualitatively in the preparation;HPLC was adopted to determine the contents of schizandrol A the main active ingredients of Wuweizi in the preparation. Results: The best extraction process of Qiongyu Runfei syrup was confirmed as the best combination: A3 B3 C1 D3, that is, soaking for four hours, adding 14 times of water,decocting for three times, three hours each time. Among them. TLC spots of Shengdihuang and Wuweizi were clear,reproducible and negative without interference;the study confirmed a method of determining the contents of schizandrol A main active ingredients of Wuweizi in the syrup, and the results showed that schizandrol A displayed the good linear relationship in the range between 0.001 04 and 0.041 6 mg/m L, r=0.9998, average recovery rate was104.56%(six cases), RSD=3.48%. Conclusion: The method established in the study, simple, accurate and stable,could be used as the standard of quality control for Qiongyu Runfei syrup.
