摘要:目的观察敦煌古医方大青龙汤(麻黄、细辛、桂枝,等)对高原低氧大鼠的肺功能、形态及血清白细胞介素-2(IL-2)、缺氧诱导因子-α(HIF-α)、转化生长因子-β(TGF-β)表达的影响作用。方法大鼠常规饲养3 d,随机分为空白组,模型组,红景天组,大青龙汤高、中、低剂量组,每组12只。各组连续灌胃7 d,从第8天开始除对照组外均进行减压低氧暴露连续14 d。末次低氧暴露后测定各组动物的肺功能。眼球取血检测血清中IL-2、HIF-α、TGF-β的含有量。取肺称重进行病理形态观察。结果与空白组比较,模型组吸气时间、呼气时间、持续时间均降低。潮气量、每分通气量、频率均升高;与模型组比较,各干预组每分通气量均降低,大青龙汤组潮气量降低,大青龙汤高剂量组、红景天组频率均降低。与空白组比较,模型组血清IL-2含有量降低,HIF-1α、TGF-β含有量均升高;与模型组比较,大青龙汤中、高剂量组,红景天组IL-2、HIF-1α含有量均升高,TGF-β含有量均降低。各组肺组织结构观察,模型组肺泡间隔显著增厚,肺毛细血管扩张,炎症细胞浸润,肺泡断裂、融合出现明显肺气肿,局部有肺大泡及肺萎陷。大青龙汤中、高剂量组,红景天组肺泡间隔增厚减轻,炎症细胞浸润减少。结论大青龙汤减轻肺损伤的防护作用可能与增强IL-2、HIF-1α表达而降低TGF-β表达有关。
AIM To observe the effects of Daqinglong Decoction( Ephedrae Herba,Asari Herba,Cinnamomi Ramulus,et al.),Dunhuang ancient medical prescription,on the lung function,pathomorphology and the expression of IL-2,HIF-1α and TGF-β in serum of altitude hypoxia rats. METHODS Conventional breeding rats for three days,the rats were randomly divided into control group,model group,Rhodiola rosea group,low-,mediumand high-dose Daqinglong Decoction groups. There were twelve rats in each group. Lavage lasted for seven days continuously. Starting from the 8th day,every rat except the control group was exposed to low pressure and low hypoxic chamber twenty-two hours after lavage thirty minutes for fourteen consecutive days. After the last hypoxic ex-posure,all the samples were taken for the following indiciators simultaneously. Lung functions were studied with animal plethysmography; blood was sampled from rat eyes and the levels of IL-2,HIF-1α,and TGF-β in serum were detected; lung was removed and weighted by electronic balance; pathological histology of lung was observed with HE staining. RESULTS Compared with the control group,indexes of inspiratory time( Ti),expiratory time( Te) and relaxation time( RT) were lowered in the model group. Compared with the model group,Ti were increased in the high-dose group. Compared with the control group,indexes of tidal volume( TV),minute ventilation( MV) and frequency were increased in the model group. Compared with the model group,MV were reduced in all interfered group; TV were reduced in Daqing long Decoction groups; frequency was slowed in the high-dose group and Rhodiola rosea group. The levels of IL-2 were lowered; however the levels of HIF-1α and TGF-β1 were increased in the serum of the model group compared with the control group. Compared with the model group,the levels of IL-2 and HIF-1 α were all elevated and TGF-β were lowered in Rhodiola rosea group medium-and highdose Daqinglong Decoction groups. The observation and comparison of lung tissue structure of all groups were carried out. The results showed that the structure of pulmonary was maintained basically complete and normal in the control group. Hypoxia lung injury in the model group was mainly featured with alveolar interval markedly thickened,pulmonary capillaries dilated,inflammatory cell infiltration and alveolar septum disrupted. As well as fusion of pulmonary alveoli,obvious pulmonary emphysema,pneumatocele and lung collapse partly presented. The pathological changes of medium-,high-dose Daqinglong Decoction and Rhodiola rosea groups lightened to different degrees as compared to the model group. CONCLUSION The protective effects of Daqinglong Decoction on pulmonary function may be related to the enhancement of the expression of IL-2 and HIF-1α and the reduction in that of TGF-β.
