摘要:目的研究七味育肝颗粒对肝纤维化模型大鼠阴虚表征和结缔组织生长因子(CTGF)表达的影响及其抗肝纤维化作用。方法将48只SPF级SD大鼠采用随机数字表法分为空白组,模型组,秋水仙碱组,七味育肝颗粒低、中、高剂量组,每组8只,采用复合方法复制肝纤维化大鼠模型,药物组给予相应药物进行干预,比较各组大鼠的进食量及饮水量增加百分数,活动、站立减少次数,舌面湿度,肛温,肝纤维化程度,肝组织CTGF平均光密度,观察各组大鼠的肝脏病理学改变。结果七味育肝颗粒可明显改善肝纤维化模型大鼠的肝脏病理改变,减轻肝组织纤维化程度(P<0.01);七味育肝颗粒可明显降低肝纤维化模型大鼠的进食量及饮水量增加百分数,增加大鼠活动及站立减少次数,升高大鼠舌面湿度,降低肛温(P<0.05或P<0.01),从而缓解肝纤维化模型大鼠的阴虚症状;七味育肝颗粒可明显降低肝纤维化模型大鼠肝组织CTGF平均光密度(P<0.05或P<0.01)。结论七味育肝颗粒对肝纤维化模型大鼠具有明显的抗肝纤维化作用,而抑制CTGF的异常高表达是其抗肝纤维化的作用机制之一;七味育肝颗粒可明显减轻肝纤维化模型大鼠的阴虚表征。
Objective To study the effects of Qiwei Yugan Keli(七味育肝颗粒,QWYGKL)on yin deficien?cy exterior syndrome and expression of connective tissue growth factor(CTGF)in model rats with hepatic fibrosis and its anti hepatic fibrosis effects.Methods Forty?eight SPF SD rats were divided into blank group,model group,colchicine group and QWYGKL low,medium and high dose groups,with 8 rats in each group.The rat model of he?patic fibrosis was replicated by compound method.The drug groups were given corresponding drugs for intervention.The rats in each group were compared in terms of the percentage increase in food and water intake,the reducing number of activities and standing,the humidity level of tongue surface,rectal temperature and the degree of liver fi?brosis,the average optical density of CTGF in liver tissue,and the liver pathological changes of rats in each group were observed.Results QWYGKL could significantly improve the pathological changes of liver in model rats with hepatic fibrosis and reduce the degree of hepatic fibrosis(P<0.01);It could significantly reduce the percentage in?crease in food and water intake,increase the reducing number of activities and standing,increase the humidity level of tongue surface and reduce the rectal temperature thereof(P<0.05 or P<0.01),thereby alleviating the yin defi?ciency syndrome thereof.It could significantly reduce the average optical density of CTGF in liver tissue of model rats with hepatic fibrosis(P<0.05 or P<0.01).Conclusion QWYGKL has marked inhibitory effect on hepatic fi?brosis,and inhibiting abnormal high expression of CTGF is one of its mechanisms.It can alleviate yin deficiency ex?terior syndrome in model rats with hepatic fibrosis.
