摘要:目的基于网络药理学方法探究冰清膏促进深Ⅱ度烧伤创面愈合的作用靶点及信号通路,探索其作用机制。方法该研究利用中药系统药理学数据库和分析平台(TCMSP)筛选冰清膏的活性成分和中药靶标,使用蛋白质数据库(Uniprot)进行基因名注释,并在基因名片数据库(GeneCards)中检索“深Ⅱ度烧伤创面愈合”的相关疾病靶点,利用Cytoscape 3.8.2软件,构建中药-活性成分-靶点网络,并采用蛋白质相互作用检索数据库(String)构建蛋白质相互作用网络(PPI),利用生物信息分析数据库(DAVID)进行潜在作用靶基因的基因功能富集分析(GO)及通路富集分析(KEGG)。结果筛选出冰清膏146个化合物和191个作用于深Ⅱ度烧伤的蛋白靶点,通过网络拓扑特征评价筛选出25个冰清膏促进深Ⅱ度烧伤创面愈合重要靶点;GO分析显示富集到生物学过程764个、细胞组分75个、分子功能149个;KEGG通路富集筛选64条主要通路在促进深Ⅱ度烧伤创面愈合方面具有作用。结论通过网络药理学预测了冰清膏促进深Ⅱ度烧伤创面愈合潜在的作用机制,为进一步的活性成分研究和实验研究提供理论依据。
Objective The targets,signaling pathways,and their mechanism of action of Bingqing Ointment in the promotion of deep second-degree burn wound healing were explored based on the network pharmacology method.Methods The active ingredients and traditional Chinese medicine targets of Bingqing Ointment were screened by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform.The annotation for gene names was performed with the protein database Uniprot.The related disease targets of"deep second-degree burn wound healing"were searched in the GeneCards database.The TCM-active ingredienttarget network was constructed by the Cytoscape 3.8.2 software.The protein-protein interaction was constructed by the protein interactions search database String.Gene ontology(GO)function enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis of the potential target genes were carried out by using the biological information annotation database DAVID.Results A total of 146 compounds of Bingqing Ointment and 191 protein targets involved in the deep second-degree burn were screened out,and 25 important targets of Bingqing Ointment for promoting deep second-degree burn wound healing were screened out based on the evaluation of network topology features.GO analysis showed that 764 biological processes,75 cellular components and 149 molecular functions were enriched.64 major pathways that function in the promotion of deep second-degree burn wound healing were screened by KEGG pathway enrichment analysis.Conclusion This study predicts the potential mechanism of action of Bingqing Ointment in the promotion of deep second-degree burn wound healing through network pharmacology,which provides a theoretical basis for further active ingredient research and experimental research.
