摘要:目的研究红芪水提物和醇提物对阿霉素(ADR)致大鼠心肌损伤的保护作用。方法给药后第8天,大鼠腹腔注射ADR(18 mg·kg^(-1))建立心肌损伤大鼠模型。按照体重将SD大鼠随机分为4组:正常组、模型组、红芪水提物组(灌胃红芪水提物10. 8 g·kg^(-1))、红芪醇提物组(红芪醇提物3. 78 g·kg^(-1)),每组10只。正常组与模型组给予20 m L·kg^(-1)的0. 9%Na Cl灌胃,1次/天,连续14 d。用酶联免疫吸附法检测血清中谷草转氨酶(AST)、肌酸激酶(CK)和乳酸脱氢酶(LDH)的含量。结果正常组、模型组、红芪水提物组、红芪醇提物组中AST的含量分别为(0. 19±0. 06),(0. 27±0. 09),(0. 19±0. 07)和(0. 17±0. 04) U·mg^(-1);这4组的LDH含量分别为(1. 47±0. 40),(1. 87±0. 40),(1. 43±0. 50)和(1. 42±0. 19) U·mg^(-1);这4组的CK的含量分别为(1. 60±0. 28),(2. 20±0. 61),(1. 53±0. 42)和(1. 38±0. 64) U·mg^(-1),模型组与正常组比较,上述指标的差异均有统计学意义(P <0. 05);红芪水提物组和红芪醇提物组与模型组比较,上述指标的差异均有统计学差义(P <0. 05,P <0. 01)。结论甘肃红芪水提物与醇提物对阿霉素所致的心肌损伤具有保护作用,红芪醇提物作用优于水提物,其作用机制与降低心肌氧化应激和提高抗氧化防御有关。
Objective To investigate the protective effect of water extract from Radix Hedysari(WRH)and ethanolic extract from Radix Hedysari(ERH)against adriamycin(ADR)-induced myocardial injury in rats.Methods A myocardial injury model induced by intraperitoneal injection of ADR was established in SD rats.ADR was administered on the 8 th day at a dose of 18 mg·kg-1 body weight.SD rats were randomly divided into four groups:normal group,model group,WRH group and ERH group,each group had ten rats.Rats in normal group and model group were orally administered with 0.9%NaCl 20 m L·kg-1,while rats in experimental groups were orally administered with WRH and ERH at the dose of 10.8 g·kg-1 and 3.78 g·kg-1,respectively.All the drugs were administered for consecutive 14 d,1 times a day.The content of glutami oxalacetic transaminase(AST),creatine kinase(CK),and lactate dehydrogenase(LDH)in the serum were determined by enzyme linked immunosorbent assay.Results The AST content of serum in normal group,model group,WRH group and ERH group were(0.19±0.06),(0.27±0.09),(0.19±0.07),(0.17±0.04)U·mg-1;the LDH content of serum in the 4 groups were(1.47±0.40),(1.87±0.40),(1.43±0.50),(1.42±0.19)U·mg-1,respectively;the CK content in the 4 groups were(1.60±0.28),(2.20±0.61),(1.53±0.42),(1.38±0.64)U·mg-1,respectively.Comparison between model group and normal group,the difference of the factors were significantly(all P<0.05);comparison between WRH group,ERH group and model group,the difference of the factors were significant(P<0.05,P<0.01).Conclusion WRH and ERH can protect against ADR-induced myocardial injury in rats,and the effect of alcohol extract is better than that of water extract.The mechanism may be related to reduce oxidative stress,enhance antioxidant defense.
