摘要:本实验旨在从Aβ代谢角度探讨当归挥发油对阿尔茨海默病(Alzheimer’s disease,AD)模型大鼠学习记忆的改善作用及其机制。大鼠分为假手术组、模型组、阳性组及当归挥发油低、中、高剂量组等6组。采用脑立体定位注射Aβ_(25-35)复制AD大鼠模型,并予以灌胃给药。Morris水迷宫检测大鼠学习记忆能力,ELISA法检测血清乙酰胆碱(acetylcholine,ACh)、胆碱乙酰转移酶(choline acetyltransferase,ChAT)活性及海马β淀粉样蛋白前体(β-amyloid precursor protein,APP)、β-淀粉样蛋白(amyloidβ-protein 1-42,Aβ_(1-42))含量,比色法测定超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、乙酰胆碱酯酶(acetylcholinesterase,AChE)水平。结果显示,当归挥发油可改善AD模型大鼠学习记忆能力,提升血清ACh、ChAT、SOD活性(P<0.05),降低血清AChE、MDA水平及海马APP、Aβ_(1-42)含量(P<0.05)。研究结果表明,当归挥发油能有效改善AD模型大鼠学习记忆能力,对AD具有一定的防治作用,其可能与调节胆碱能神经递质、自由基氧化等介导的Aβ代谢有关。
This study aims to explore the effect and mechanism of Angelica volatile oil on learning and memory in Alzheimer’s disease(AD)model rats.The rats were divided into 6 groups:sham operation group,model group,positive group and low,medium and high dose groups of Angelica volatile oil.Brain stereotactic injection of Aβ_(25-35) was used to establish AD rat model and the drugs were administered by gavage.The learning and memory ability of rats was examined by Morris water maze.Serum acetylcholine(ACh),activity of choline acetyltransferase(ChAT)andβ-amyloid precursor protein(APP),amyloidβ-protein 1-42(Aβ_(1-42))in hippocampus were detected by ELISA.The levels of superoxide dismutase(SOD),malondialdehyde(MDA)and acetylcholinesterase(AChE)were measured by colorimetry.The results showed that Angelica volatile oil could improve the learning and memory ability of AD model rats,increase the activities of Ach,chat and SOD(P<0.05),and reduce serum AChE,the level of MDA and content of hippocampus APP and Aβ_(1-42)(P<0.05).The results show that Angelica volatile oil can effectively improve the learning and memory ability of AD model rats and has preventive and therapeutic effect on AD,which may be related to the regulation of cholinergic neurotransmitter,free radical oxidation and Aβmetabolism.
