摘要:目的探讨黄芪甲苷通过TGF-β/Smad2信号通路对心力衰竭(HF)大鼠心肌纤维化(MF)的影响。方法采用腹腔注射2.5 mg·kg^(-1)阿霉素,1次/周,连续6周的方法造模。将40只雄性Wistar大鼠随机分为对照组、模型组、盐酸贝那普利组(1.1 mg^(-1)·kg·d^(-1))、黄芪甲苷组(80 mg·kg^(-1)·d^(-1)),每组10只,造模成功后干预6周,末次给药24 h末采用心脏彩色多普勒超声评价心功能,Western Blot检测大鼠心脏组织转化生长因子-β(TGF-β)、受体调节的分子2(Smad2)的表达,采用免疫荧光检测大鼠心肌组织基质金属蛋白酶(MMP)-9、α-平滑肌肌动蛋白(SMA)的表达,Masson染色检测大鼠心肌组织心肌纤维化程度。结果与对照组比较,模型组LVEF、LVFS升高(P<0.05),TGF-β、Smad2、MMP-9、α-SMA蛋白表达升高(P<0.05),纤维化程度严重;与模型组比较,盐酸贝那普利组、黄芪甲苷组LVEF、LVFS降低(P<0.05),TGF-β、Smad2、MMP-9、α-SMA蛋白表达降低(P<0.05),纤维化程度减轻。结论黄芪甲苷可能通过TGF-β/Smad2信号通路对阿霉素诱导的心力衰竭大鼠心肌纤维化具有一定的缓解作用。
Objective To investigate the effects of astragaloside on myocardial fibrosis(MF)in rats with heart failure(HF)through TGF-β/Smad2 signaling pathway.Methods The model was made by intraperitoneal injection of 2.5 mg·kg^(-1)adriamy-cin once a week for 6 weeks.Forty male Wistar rats were randomly divided into normal group,model group,Benazepril hydrochlo-ride group(1.1 mg·kg^(-1)·d^(-1))and astragaloside side group(80 mg·kg^(-1)·d^(-1)),with 10 rats in each group.The interven-tion lasted for 6 weeks after successful modeling,and cardiac function was evaluated by color Doppler ultrasound at the end of the last administration 24 hours.The expressions of transforming growth factor-β(TGF-β)and receptor-regulated molecule 2(Smad2)in rat heart tissue were detected by Western Blot,and the expressions of matrix metalloproteinase(MMP)-9 andα-smooth muscle actin(SMA)were detected by immunofluorescence.The degree of myocardial fibrosis in rat myocardium was de-tected by Masson staining.Results Compared with normal group,LVEF and LVFS in model group were increased(P<0.05),TGF-β,Smad2,MMP-9 andα-SMA protein expression were increased(P<0.05),and the degree of fibrosis was serious.Compared with model group,LVEF and LVFS in Benazepril hydrochloride group and Astragaloside group were decreased(P<0.05),the protein expressions of TGF-β,Smad2,MMP-9 andα-SMA were decreased(P<0.05),and the degree of fibro-sis was alleviated.Conclusion:Astragaloside may alleviate doxorubicin-induced myocardial fibrosis in rats with heart failure through TGF-β/Smad2 signaling pathway.
