摘要：目的:探讨脾肾阳虚型溃疡性结肠炎(Ulcerative Colitis,UC)大鼠中基质金属蛋白酶(Matrix Metalloproteinase,MMP)3、MMP8、MMP13的特异性。方法:96只Wistar大鼠随机分为空白组、模型组、理中汤合四神丸复方颗粒高剂量组、中剂量组、低剂量组、柳氮磺胺吡啶(SASP)组,治疗组给予相应药物灌胃治疗。利用高通量测序技术筛选出差异表达的MMP3、MMP8、MMP13,进一步用实时荧光定量PCR(RT-q PCR)法和蛋白质印迹法(Western blot)法验证其表达。结果:与空白组相比较,模型组结肠组织中MMP3、MMP8、MMP13基因与蛋白表达均上调(P<0.01);与模型组相比较,各治疗组结肠组织MMP3、MMP8、MMP13基因与蛋白表达均下调(P<0.01)。结论:具有温补脾肾作用的理中汤合四神丸复方颗粒能显著下调脾肾阳虚型UC大鼠结肠黏膜中过度表达的MMP3、MMP8、MMP13基因和蛋白,抑制炎症反应及黏膜的破坏,亦可作为诊断UC的特异性标志物及治疗UC的疗效评估指标。
Objective:To verify the specificity of MMP3,MMP8 and MMP13 in UC rats with the Pishen Yangxu type(脾肾阳虚型).Methods:96 Wistar rats were randomly divided into the control group,model group,high dose group,middle dose group,low dose group and SASP group.The treatment group was treated with corresponding drugs by gastric perfusion.The differentially expressed MMP3,MMP8,MMP13 were screened by high throughput sequencing,and further verified by RT-q PCR and Western blot.Results:Compared with the control group,the expression of MMP3,MMP8,MMP13 gene and protein were up-regulated in colonic tissue of model group(P<0.01).Compared with the model group,the expression of MMP3,MMP8,MMP13 gene and protein were down-regulated in colonic tissue of model group(P<0.01).Conclusion:The granules of Lizhong decoction and Sishen Wan(理中汤合四神丸复方颗粒)showed the effect on Wenbu Pishen(温补脾肾),can significantly down-regulate the overexpression of MMP3,MMP8,MMP13 gene and protein in colonic mucosa of UC rats with the Pishen Yangxu type,and inhibit inflammatory reaction and mucosal damage.It also can be used as a specific marker for the diagnosis of UC and an evaluation index for the therapeutic effect on UC.
