摘要:目的利用分子生物学技术探讨针刺三焦经腧穴防治噪音性耳聋的分子机制。方法雄性Wistar大鼠随机分成空白对照组、噪音性耳聋模型组、维生素E治疗组和针刺治疗组。针刺组取三焦经腧穴"翳风"(双)、"外关"(双)。各组大鼠在实验结束后进行听觉脑干诱发电位(ABR)和耳蜗毛细胞VEGF免疫组织化学检测。结果噪音性耳聋模型组和空白组相比,大鼠ABR阈值异常升高,差异有极其显著性意义(P<0.01),针刺治疗组、维E治疗组和模型组相比,大鼠ABR阈值显著改善,差异有极其显著性意义(P<0.01),并且针刺治疗组优于维E治疗组(P<0.05);噪音性耳聋模型组和空白组相比,大鼠耳蜗基底膜毛细胞VEGF免疫阳性细胞数量明显降低,差异有显著性意义(P<0.05);针刺治疗组、维E治疗组和模型组相比,大鼠耳蜗基底膜毛细胞VEGF免疫阳性细胞显著恢复,差异有显著性意义(P<0.01),但针刺治疗组和维E治疗组相比,差异不显著。结论长期噪声刺激可使大鼠听敏度下降,听阈升高,产生永久性阈移(PTS),针刺和维生素E治疗均能降低ABR的阈值,改善噪声导致的听力损害,针刺的效果优于维生素E;VEGF在治疗组大鼠耳蜗毛细胞中的高表达,可能是通过治疗增加了耳蜗新生血管的形成改善了耳蜗供氧所致。
Objective Using molecular biology techniques to explore the molecular mechanism of acupuncture San - Jiao points control noise - induced hearing loss in rats. Methods Adult Wistar rats are randomly divided into blank control group, noise - induced hearing loss rats model group, acupuncture treatment group, vitamin E treatment group. To get the “Yifeng”and “WaiGuan” of the Acupuncture group. Rats in each group at the end of the experiment after the auditory brainstem response (ABR) testing and cochlear hair cell VEGF immunohistochemistry check to basilar membrane stretched preparation. Results Noise - induced hearing loss rats model group compares with blank control group,auditory brainstem response (ABR) threshold of the rats anomalies increased ,the difference is highly significant (P 〈0. O1 ). Acupuncture treatment group and Vitamin E treatment group compares with model group, auditory brainstem response (ABR) threshold of the rats is significantly improved, the difference is highly significant ( P 〈0.01 ), and the acupuncture treatment group is better than Vitamin E treatment group ( P 〈0.05 ). Noise - induced hearing loss rats model group compared with the blank control group, the immunoreactive cells of rat cochlear hair cells in VEGF are significantly decreased, the difference is significant (P 〈0.05 ) ;acupuncture treatment group and Vitamin E treatment group compared with model group, the immunoreactive cells of rats cochlear hair cells in VEGF are significantly restored, the difference is significant (P 〈0.01 ), but the two groups had no significant difference in results. Conclusion The long- term noise stimulation can result in significant harm to the hearing of rats, acupuncture and Vitamin E treatment can improve the noise - induced hearing damage, and the effect of acupuncture is superior to Vitamin E. The long - term noise stimulation can lead to reduce the expression of VEGF in cochlear hair and decrease blood supply. Acupuncture and Vitamin E treatment can promote the expression of VEGF in cochlear hair and improve hearing of rats, but both acupuncture and vitamin E treatment have no significant difference.
