摘要:目的:研究地黄饮子水煎剂对去势大鼠生物力学和OPG/RANKL/RANK含量的影响,进一步探讨地黄饮子防治绝经后骨质疏松症的作用机制。方法:采用去卵巢法制备大鼠骨质疏松症模型,将大鼠随机分为假手术组,模型组,地黄饮子水煎剂低、中、高剂量组,西药对照组(戊酸雌二醇片),每组10只,分别予相应药物灌胃4周,股动脉采血后处死大鼠。计算各组大鼠的胸腺、脾脏和肾脏指数,ELISA试剂盒检测血清OPG、RANK和RANKL的含量;双能X线骨密度仪测量获得骨矿含量(BMC);AG-IX生物力学万能实验机检测断裂载荷和弹性模量。结果:与假手术组比较,模型组大鼠血清RANK和RANKL含量均升高(P﹤0.05),胸腺、脾脏、肾脏指数及血清OPG含量、BMC、生物力学参数断裂载荷、弹性模量均明显降低(P﹤0.05)。与模型组比较,各用药组大鼠血清RANK和RANKL含量均降低(P﹤0.05),胸腺、脾脏指数、OPG含量、BMC、断裂载荷和弹性模量均显著升高(P﹤0.05)。结论:地黄饮子具有明显抗去卵巢大鼠骨质疏松作用,改善骨组织中生物力学参数和调控OPG/RANKL/RANK信号通路是其可能作用机制。
Objective: To study the effects of Dihuang Yinzi Decoction on biomechanics and OPG/RANKL/RANK content of ovariectomized rats and further discuss the mechanism of Dihuang Yinzi Decoction in preventing and treating postmenopausal osteoporosis (PMOP). Methods: Osteoporosis rat model was established by ovariectomized method, the rats were randomly divided into sham operation group, model group, Dihuang Yinzi Decoction high, medium, low dose group, western medicine control group (estradiol valerate tablets), with 10 rats in each group. The rats were gavaged with corresponding drugs for 4 weeks and killed after their blood were collected from the femoral artery. The indexes of thymus, spleen and kidney were calculated, and the contents of OPG, RANKL and RANK in serum were detected by enzyme-linked immune sorbent assay (ELISA) kits, bone mineral content (BMC) was examined with dual energy absorptiometer, fracture load and elasticity modulust were determined with AG-IX biomechanical universal testing machine. Results: Compared with the sham operation group, the contents of RANK and RANKL were both increased in model control group (P〈0.05), while the indexes of thymus, spleen kidney and the serum OPG content, BMC, the fracture load and elastic modulus of the biomechanical parameters were significantly reduced in modern group (P〈0.05). Compared with the model group, the contents of RANK and RANKL were both markedly decreased in each Dihuang Yinzi Decoction intervention group (P〈0.05). However, the indexes of spleen and thymus, OPG content, BMC, fracture load and elasticity modulust were all significantly increased in Dihuang Yinzi Decoction intervention group (P〈0.05). Conclusion: Dihuang Yinzi Decoction has obvious anti-osteoporosis effect in ovariectomized rats, and the improvement of biomechanical parameters in bone tissue and the regulation of OPG/RANKL/RANK signaling pathway are its possible mechanisms.
