摘要:目的比较小鼠口服甘草汁蒸制高乌头不同溶剂萃取部分的急性毒性。方法采用系统溶剂法,对甘草汁制高乌头95%乙醇提取物进行分离,依次得到石油醚、氯仿、乙酸乙酯、正丁醇、乙醇和水提6个不同的溶剂萃取部分,通过测定小鼠口服半数致死量(LD_(50))和最大给药量,对分离得到的不同溶剂萃取部分进行急性毒性比较。结果甘草汁制高乌头三氯甲烷、正丁醇及水提部分的LD_(50)分别为114.10,870.48,2422.22 mg·kg^(-1),95%可信限分别为100.70~126.86,487.59~1072.23,2032.98~2841.73 mg·kg^(-1);乙酸乙酯、石油醚及乙醇部分最大给药量分别为1976.10,13116.00,20316.00 mg·kg^(-1)。结论高乌头生品有毒,经甘草汁蒸制后仍具一定毒性,三氯甲烷部分是制高乌头的"毒性部位群",毒性成分主要存在于三氯甲的烷部分。
Objective To compare the oral toxicity in mice of different extraction fractions of steamed Aconitum sinomontanum Nakai by licorice.Methods The experimental drugs were obtained by the means of systematic solvent extraction from the 95% ethanol extract. The oral acute toxicity in mice of six different extraction fractions( petroleum ether,chloroform,ethyl acetate,butanol,alcohol and water) were carried out by the way of measuring the median lethal dose( LD_(50)) and maximum dose. Results LD_(50) of the chloroform,butanol and water extractions were 114. 10,870. 48 and 2422. 22 mg · kg-1. The 95% confidence limits were 100. 70- 126. 86, 487. 59- 1072. 23 and 2032. 98-2841. 73 mg·kg-1,respectively. The maximum dose of the ethyl acetate,petroleum ether and alcohol extractions were 1976. 10,13116. 00 and 20316. 00 mg·kg-1,respectively. Conclusion The raw Aconitum sinomontanum Nakai is toxic,and the steamed pro-ducts are still toxic.The chloroform section is the toxic fraction group to the processed products. The toxic components of the processed Aconitum sinomontanum Nakai mainly exist in the chloroform fraction.
