摘要:【目的】探讨秦巴硒菇多糖(FA-2-b-β)联合盐酸表阿霉素(Epirubicin Hydrochloride,EPI)对骨肉瘤MG63细胞增殖、迁移及凋亡的影响。【方法】采用MTT法测定FA-2-b-β、EPI和二者联合用药时对MG63细胞的抑制率,并根据金氏公式判断两药的协同效应,同时在倒置显微镜下观察细胞形态;划痕实验检测细胞增殖;DAPI染色及流式细胞术检测细胞凋亡;JC-1染色检测线粒体膜电位的变化;蛋白质免疫印迹(Western Blot,WB)和qPCR检测各组细胞PI3K、AKT、mTOR、Bax、Bcl-2、Capase-3的表达。【结果】FA-2-b-β、EPI及二者联合用药对MG63细胞的增殖均有抑制作用,联合用药组的增殖抑制作用更显著,具有协同效应;倒置显微镜可见联合用药组存活细胞数量减少,体积变大;与单一用药相比,联合用药时诱导MG63细胞凋亡、抑制细胞迁移和降低细胞线粒体膜电位的作用均更显著(P<0.05);FA-2-b-β、EPI和二者联用给药均可上调Bax、Capase-3的表达(P<0.05),下调Bcl-2、PI3K、AKT、mTOR的表达(P<0.05),联合用药组的作用更为显著(P<0.05)。【结论】FA-2-b-β联合EPI可协同抑制骨肉瘤MG63的增殖和迁移,并诱导凋亡,该作用可能与负向调控骨肉瘤MG63细胞中的PI3K/AKT/mTOR信号通路有关。
【Objective】To investigate the effect of Agaricus blazei Murrill polysaccharide(FA-2-b-β)combined with epirubicin(EPI)on the migration and apoptosis of osteosarcoma MG63 cells.【Method】The effects of FA-2-b-βand EPI on the proliferation of MG63 cells were determined by MTT colorimetric method,the synergistic effect of the two drugs was judged according to King's formula,and cell morphology was observed under an inverted microscope;scratch experiments to detect cell proliferation;DAPI staining and flow cytometry to detect apoptosis;JC-1 staining was used to detect the change of mitochondrial mem-brane potential;qPCR to detect the expression of PI3K,AKT,mTOR,Bax,Bcl-2,Capase-3 genes in each group;detection of the expression of PI3K,AKT,mTOR,Bax,Bcl-2,Capase-3 protein by Western blot.【Result】FA-2-b-βand EPI could significantly inhibit the proliferation of MG63 cells in a dose-dependent manner,the inhibitory effect of the combined drug group was more significant and had a synergistic effect.The inverted microscope showed that the number of surviving cells in the combined drug treatment de-creased and the volume increased.The combined administration of FA-2-b-βand EPI could significantly in-duce apoptosis of MG63 cells,inhibit cell migration,and reduce the mitochondrial membrane potential of cells.FA-2-b-β,EPI and combined administration all groups upregulated the expressions of Bax and Ca-pase-3(P<0.05)and downregulated the expressions of Bcl-2,PI3K,AKT and mTOR(P<0.05),and the effect of the combined administration group was more significant(P<0.05).【Conclusion】FA-2-b-βcom-bined with EPI can synergistically inhibit the proliferation and migration of osteosarcoma MG63 cells and in-duce apoptosis,which may be related to the negative regulation of PI3K/AKT/mTOR signaling pathway in osteosarcoma MG63 cells.
