摘要:目的:研究敦煌医方四时常服方对镉染毒大鼠脑氧化应激功能的影响。方法:选取Wistar大鼠60只,随机分为6组,分别为空白对照组,阳性药物对照组,单纯镉染毒模型组,四时常服方低剂量干预组、中剂量干预组和高剂量干预组。对大鼠镉染毒的同时进行药物干预治疗,五周后处死大鼠,取脑组织及动脉血。分别测定大鼠脑组织中超氧化物歧化酶(SOD)和丙二醛(MDA)含量以及血清中乳酸脱氢酶(LDH)含量。结果:与空白对照组比较,单纯镉染毒模型组脑组织SOD含量明显降低(P<0.01),MDA含量明显升高(P<0.01);与模型组比较,四时常服方药物组脑组织SOD含量显著升高(P<0.01),MDA含量显著降低(P<0.01),以高剂量组最为明显,差异具有统计学意义。此外,与空白对照组比较,模型组血清中LDH含量明显上升(P<0.01);与模型组比较,四时常服方药物组血清中LDH含量显著下降(P<0.01),以高剂量组最为明显,差异具有统计学意义。结论:四时常服方对于镉染毒大鼠脑氧化应激功能有一定影响。
Objective:To research the effect of the medical prescription of Sishi Changfu on oxidative stress in rats induced by cadmium. Methods:60 Wistar rats were divided into 6 groups randomly which includes healthy-control group, positive drug-control group, pure cadmium exposure-model group,and low dose drug-intervention group,medium dose drug-intervention group and high dose drug-intervention group. The rats were treated with drug intervention at the same time of cadmium exposure. Five weeks later, the rats were killed and the brain tissue and arterial blood were taken. The level of superoxide dismutase (SOD) and malondialdehyde (MDA) in rat brain tissue were measured, then the content of superoxide dismutase (SOD), malondialdehyde(MDA), glutathione peroxidase(GSH-Px), lactate deh- ydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum were also measured. Results: Compared with the healthy-control group,the brain tissue SOD level decreased significantly(P〈0.01 ) and the contents of MDA increased significantly(P〈0.01 ) in pure cadmium exposure-model group; For drug-intervention groups, the level of SOD were significantly increased (P〈0.01) and the content of MDA decreased significantly(P〈0.01 ) in brain tissue compared with the cadmium exposure-model group, the difference which was most obvious in high dose-drug group. In addition, compared with the healthy-control group,the content of LDH in serum was significantly increased (P〈0.01) in model group; Compared with the model group, the drug intervention groups has lower content of LDH (P〈0.01), the difference was statistically significant especially in high dose-drug group. Conclusion:the medical prescription of sishichangfu has certain effect on the recovery of renal function and cerebral oxidative stress in rats exposed to cadmium.
