摘要:Sema(semaphorin)家族作为经典轴突导向因子,近年被证实在成血管-成骨耦联中发挥重要作用,通过与神经纤毛蛋白1/神经纤毛蛋白2(neuropilin1/neuropilin2, NRP1/NRP2)及Plexin受体(Plexin, A~D型)互作,不仅影响内皮细胞迁移与血管分支,还调控成骨细胞与破骨细胞的动态平衡,并与多条信号通路发生交叉,不同Sema成员在成血管-成骨微环境中具有差异化的功能。该文系统综述Sema家族在成骨–成血管耦联中的分子机制与研究进展,并提出结合单细胞/空间组学与生物材料递送策略的未来方向,以期为骨相关疾病的精准治疗提供新靶点与新思路。
The Sema(semaphorin)family,traditionally recognized as classic axon guidance molecules,has recently been shown to play pivotal roles in angiogenesis-osteogenesis coupling.By interacting with NRP1/NRP2(neuropilin1/neuropilin2)and Plexin-A-Plexin-D,semaphorins not only regulate endothelial cell migration and vascular branching but also modulate the dynamic balance between osteoblasts and osteoclasts.Moreover,they engage in crosstalk with multiple signaling pathways,with distinct Sema members exhibiting differential functions within the angiogenicosteogenic microenvironment.This review systematically summarizes the molecular mechanisms and recent advances concerning the role of the Sema family in angiogenesis-osteogenesis coupling.Furthermore,future research should integrate single-cell/spatial omics and biomaterial-based delivery strategies,aiming to provide novel targets and innovative approaches for the precision treatment of bone-related diseases.
