摘要:目的:研究参苓白术散对脾虚湿困型溃疡性结肠炎大鼠结肠组织p38MAPK、TNF-α及IL-4基因、蛋白表达的干预作用。方法:SPF级Wistar大鼠60只,按随机数字表法标记,按性别不同分笼饲养。取雌、雄大鼠各5只作为空白组,其余大鼠依据性别不同分笼饲养;采用复合因素+TNBS/乙醇刺激法(乙醇浓度50%)构建脾虚湿困型UC大鼠模型;造模成功后,依随机数字表法将模型大鼠分为模型组、参苓白术散低、中、高治疗组,剂量分别为6 g/(kg·d)、12 g/(kg·d)、24 g/(kg·d)及阳性对照组,柳氮磺胺吡啶组,SASP组0. 2 g/(kg·d),每组各10只,雌雄各半、分笼饲养。其余各组大鼠均以蒸馏水灌胃治疗,剂量:10 g/(kg·d),疗程3周,每日灌胃1次。免疫组化法检测各组实验大鼠结肠组织p38MAPK、TNF-α、IL-4的蛋白含量表达; RT-PCR法检测UC各组大鼠结肠组织内p38MAPK、TNF-α及IL-4的基因表达水平。结果:与空白组比较,模型组大鼠结肠组织内p38MAPK、TNF-α蛋白含量、基因表达水平明显升高(P <0. 01或P <0. 05); IL-4的含量蛋白、基因表达水平显著降低(P <0. 01或P <0. 05);与模型组相比较,阳性对照组、参苓白术散中、高剂量组大鼠p38MAPK、TNF-α基因、蛋白表达水平降低显著(P <0. 01或P <0. 05); IL-4的蛋白含量、基因表达水平明显升高(P <0. 01或P <0. 05),尤以参苓白术散高剂量组及阳性对照组作用显著。结论:参苓白术散可明显调节脾虚湿困型UC大鼠病变部位结肠组织内p38MAPK、TNF-α及IL-4基因及蛋白表达的水平。
Objective: To study the effect of Shenling Baizhu Powder on colonic protein and mRNA expressions of p38MAPK, TNF -α and IL -4 in rats with ulcerative colitis(UC) of the pattern of Spleen deficiency and dampness re- tention. Methods: 60 SPF Wistar rats were assigned randomly and fed in different groups according to gender. Five fe- male rats and five male rats were assigned to the blank group; others were fed in different cages according to gender. The UC model was established by combination method (TNBS/50% ethanol). The successful models were randomly di- vided into the model group, Shenling Baizhu Powder groups(6 g/kg · d, 12 g/kg · d, 24 g/kg · d), and SASP group (sulfasalazine, 0.2 g/kg · d) ; with 10 rats in each group. Rats in the blank and the model group were intragastrically with distilled water (10 g/kg ·d ). The treatment course was thine weeks. The protein and mRNA expessions of p38MAPK, TNF-α and IL-4 in colonic tissues were detected by inmmnohistochemistry and RT- PCR respectively. Results: Compared to the control group, the protein and mRNA expressions of p38MAPK and TNF- α were significantly increased in the model group(P 〈 0.01, P 〈 0.05) ; the protein and mRNA expressions of IL- 4 were significantly de- creased (P 〈 0.01, P 〈 0.05). Compaared to the model group, protein and mRNA expressions of p38MAPK and TNF - αwere significantly decreased in SASP group and in Shenling Baizhu Powder roups of medium - dose and high - dose (P 〈 0.01, P 〈 0.05) ; the protein and mRNA expressions of IL -4 were significantly increased in the SASP group and Shengling Baizhu Powder group of high - dose(P 〈 0.01, P 〈 0.05 ). Conclusion : Shenling Baizhu Powder can signifi- cantly regulate the protein and mRNA expressions of p38MAPK, TNF -αand IL - 4 in UC rats with the pattern of Spleen deficiency and dampness retention.
